摘要:
目的探讨高良姜、大高良姜、红豆蔻3味山姜属中药黄酮对胃溃疡寒证大鼠胃消化液的影响及作用机制。 方法选择90只SPF级SD大鼠,随机抽取10只大鼠为空白组,其余大鼠进行胃溃疡寒证造模,末次造模结束后随机分为模型组,干姜姜辣素组,高良姜高、低剂量组,大高良姜高、低剂量组,红豆蔻高、低剂量组,共9组。采用灌服冰知母水煎液与15%冰乙酸制备大鼠胃溃疡寒证模型,以干姜姜辣素为阳性对照,测量大鼠胃液量、胃酸pH值,ELISA法测定胃蛋白酶活力、胃动素(motilin,MTL)、胃泌素(gastrin,GAS)、生长抑制素(somatostatin,SS)含量及H+-K+-ATP酶活力。 结果与空白组大鼠比较,模型组大鼠胃液量、胃蛋白酶活力、GAS、MTL含量、H+-K+-ATP酶活力均升高(P<0.01);胃酸pH值,SS含量显著减少(P<0.01)。与模型组比较,干姜姜辣素组,高良姜、大高良姜、红豆蔻高剂量组大鼠胃液量、胃蛋白酶活力显著降低(P<0.01或P<0.05),pH显著升高(P<0.01或P<0.05);高良姜、大高良姜低剂量组胃液量显著减少(P<0.01或P<0.05),pH显著升高(P<0.01或P<0.05);干姜姜辣素组,高良姜高低剂量组,大高良姜、红豆蔻高剂量组大鼠胃组织GAS含量、MTL含量、H+-K+-ATP酶活力显著降低(P<0.01或P<0.05),SS含量显著增加(P<0.01或P<0.05)。 结论3味山姜属中药黄酮类成分能够抑制胃溃疡寒证大鼠胃液分泌、提高胃酸pH值、降低胃蛋白酶活力,其作用机制与减少GAS、MTL含量、增加SS含量、减少H+-K+-ATP酶活力有关,推测其通过对胃肠活动的影响,减少胃液分泌以及胃肠活动,通过对质子泵的影响,抑制H+涌入胃腔,降低胃酸的生成,维持胃黏膜细胞内外离子平衡。
Abstract:
ObjectiveTo discuss the effects of the flavonoids extracted from Gaoliangjiang (Alpiniae Officinarum Rhizoma), Da Gaoliangjiang (Alpinia galanga Swartz) and Hongdoukou (Galangae Fructus) on gastric digestive fluid secretion in rats with cold pattern gastric ulcer and the mechanism. MethodsAll 90 SPF grade SD rats were chosen, ten ones were randomly selected as the blank group, the rest were prepared into the models with cold pattern gastric ulcer, after the last modeling, they were randomized into the model group, dried gingerol group, high and low dosages groups of Gaoliangjiang, high and low dosages groups of Da Gaoliangjiang, and high and low dosages groups of Hongdoukou, nine groups altogether. Gavage administration of frozen decoction of Zhimu (Anemarrhenae Rhizoma) and 15% glacial acetic acid were adopted to build rat models with cold pattern gastric ulcer, dried gingerol was chosen as negative control, to detect the amount of gastric juice and pH value of gastric juice of the rats, ELISA method was used to measure stomach prolease activity, the contents of MTL, GAS and SS, as well as the activity of H+-K+-ATP enzyme. ResultsCompared with the blank group, the amount of gastric juice, the activity of stomach prolease, the contents of GAS, MTL and the activity of H+-K+-ATP enzyme raised in the model group (P<0.05); pH value of gastric juice and the contents of SS were reduced remarkably (P<0.01). Compared with the model group, the amount of gastric juice and the activity of stomach prolease were remarkably lowered in dried gingerol group, high doses groups of Gaoliangjiang, Da Gaoliangjiang and Hongdoukou (P<0.01 or P<0.05), pH was significantly increased (P<0.01 or P<0.05); the amount of gastric juice was noticeably reduced in low doses groups of Gaoliangjiang and Da Gaoliangjiang (P<0.01 or P<0.05), pH was significantly increased (P<0.01 or P<0.05); the contents of MTL and GAS, the activity of H+-K+-ATP enzyme was significantly reduced in dried gingerol group, high and low doses groups of Gaoliangjiang, high doses groups of Da Gaoliangjiang and Hongdoukou (P<0.01 or P<0.05); the contents of SS was significantly increased (P<0.01 or P<0.05). ConclusionThe flavonoids extracted from Gaoliangjiang, Da Gaoliangjiang and Hongdoukou could inhibit the secretion of gastric juice in rat with cold pattern gastric ulcer, raise pH of gastric juice, lower the activity of stomach prolease, and its mechanism might be related to reducing the contents of MTL and GAS, increasing the contents of SS, decreasing H+-K+-ATP enzyme, it is speculated that it could decrease the secretion of gastric juice and gastrointestinal activity by its influence on gastrointestinal activity, it could inhibit the reflux of H+ into the gastric cavity, reduce the production of stomach acid and maintain the internal and external ion balance of gastric mucosal cells through its impacts on proton pump.
