摘要:
目的研究黄金胶囊对2型糖尿病大鼠内质网应激因子肌醇需求酶1(inositol-requiring enzyme-1,IRE-1)、转录因子C/EBP同源蛋白(C/EBP-homologous protein,CHOP)表达的作用,进一步挖掘黄金胶囊深层次转变胰岛功能的机理。 方法首先将SPF雄性Wistar大鼠按造模与否随机分组:空白组(8只)和造模组(48只)。在高脂高糖饲料诱导下应用小剂量链脲佐菌素(streptozotocin,STZ)损害胰岛细胞,建立2型糖尿病(type 2 diabetic mellitus,T2DM)大鼠模型,将造模成功的40只大鼠随机分为:模型组,黄金胶囊低、中、高剂量组,罗格列酮组,每组8只。各组给予对应药物灌胃治疗,空白组与模型组给予生理盐水灌胃,共连续进行灌胃10周后处死各组大鼠。观察各组大鼠血糖指标、血脂指标水平。聚合酶链反应与免疫印迹测定实验大鼠肝组织样本中IRE-1、CHOP mRNA及蛋白分子表达变化,HE染色观察肝组织病理形态学变化。 结果与模型组比较,罗格列酮组及黄金胶囊组血空腹血糖(fasting blood glucose,FBG)及血脂水平均下降(P<0.05);与常规罗格列酮组比较,黄金胶囊中、低剂量组疗效欠佳(P<0.05),然而高剂量组与之功效接近(P>0.05)。与空白组比较,模型组样本中IRE-1、CHOP的蛋白分子表达和mRNA的转录增加(P<0.05);与模型组比较,罗格列酮组和黄金胶囊组IRE-1、CHOP mRNA表达均降低(P<0.05);与罗格列酮组比较,黄金胶囊中药中、低剂量组作用不及罗格列酮组(P<0.05),而中药高剂量组与之相近(P>0.05)。HE染色显示采用黄金胶囊灌胃大鼠的肝组织病理炎性反应下降,尤其在高剂量组突出。 结论黄金胶囊可通过减少肝组织中IRE-1、CHOP表达,改善组织病理损害,最终缓解肝脏内质网应激,调控血糖、血脂。
Abstract:
ObjectiveTo study the effects of Huangjin capsules on the expressions of the ER stress factors IRE-1 and CHOP in rats with type two diabetes mellitus (T2DM), so as to further unearth the in-depth mechanism of Huangjin capsules transforming islet function. MethodsSPF male Wistar rats were allocated to blank group (eight rats) and modeling group (48 rats) according to whether they accept modeling or not. Induced by high-fat and high-sugar feed, islet cells were damaged by using small dose of STZ, to establish T2DM rat models, after successful modeling, 40 rat models were randomized into the model group, low, moderate and high dose groups of Huangjin capsules, rosiglitazone group, eight rats each group. Different groups were drenched with the correspon-ding medicine, the blank group and the model group were perfused with physological saline, the rats in all the groups were sacrificed after gastric gavage for ten weeks consecutively. To observe the levels of blood glucose and blood lipid of the rats in different groups. Polymerase chain reaction and Western Blotting were used to detect the expressions of IRE-1, CHOP mRNA and protein molecules in liver tissue of the rats, HE staining to observe the pathomorphology of liver tissue. ResultsCompared with the model group, the levels of FBG and blood lipid reduced in rosiglitazone group and Huangjin capsules group (P<0.05). Compared with conventional rosiglitazone group, clinical effects of moderate and low dose groups of Huangjin capsules were unsatisfactory (P<0.05), however, close to clinical effects of high dose group (P>0.05). Compared with the blank group, the expressions of IRE-1 and CHOP protein molecules and mRNA transcription increased in the model group (P<0.05); compared with the model group, the expressions of IRE-1 and CHOP mRNA lowered in rosiglitazone group and Huangjin capsules group (P<0.05); moderate and low dose groups of Huangjin capsules were less effective than rosiglitazone group (P<0.05), while its clinical effects close to high dose group of herbs (P>0.05). HE staining displayed that pathological inflammatory response of the rats by gastric gavage of Huangjin capsules reduced, especially noticeable in high dose group. ConclusionHuangjin capsules could finally relieve liver endoplasmic reticulum stress and regulate blood glucose and blood lipid by reducing the expressions of IRE-1 and CHOP in liver tissue and improving histopathological damage.
