摘要:
目的通过网络药理学方法探讨海桐皮治疗类风湿关节炎(rheumatoid arthritis,RA)的作用机制。 方法在中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)检索海桐皮有效成分及其相关靶点,并利用Perl脚本对海桐皮相关靶点进行基因注释后获取海桐皮的相关靶点基因。分别在人类基因数据库(the human gene database,GeneCards)、Disgenet数据库检索RA疾病基因,利用R×64 3.5.3软件对药物相关靶点基因和疾病基因进行交互,取得药物-疾病之间的交集基因;运用Cytoscape 3.7.1软件构建药物-有效成分-疾病-基因网络;利用String在线网站对交集基因进行蛋白互作网络构建;最后运用R×64 3.5.3软件及Perl脚本对交集基因进行基因本体论功能富集分析(gene ontology,GO)和KEGG富集分析(kyoto encyclopedia of genes and genomes,KEGG)。 结果在TCMSP数据库检索到海桐皮有效成分6个,相关靶点225个,海桐皮与RA交集基因14个;海桐皮治疗RA主要涉及的生物过程包括肾上腺素受体活性、G蛋白偶联的胺受体活性、儿茶酚胺结合、肾上腺素受体结合等过程;主要涉及的信号通路包括cGMP-PKG信号通路、神经活性配体-受体相互作用信号通路及cAMP等信号通路等。 结论海桐皮治疗RA通过多成分、多靶点、多通路发挥作用,但具体作用机制尚需要动物实验与临床试验的进一步验证。
Abstract:
ObjectiveTo explore the mechanism of Haitongpi in the treatment of rheumatoid arthritis (RA) via network pharmacology. MethodsThe active ingredients and its related targets of Haitongpi were searched from TCMSP, genetic annotation of Haitongpi-related targets was conducted using Perl scripts, to obtain its related target genes. RA disease genes were retrieved from GeneCards and Disgenet respectively, the genes were interacted with drug-related target genes using R×64 3.5.3 software to obtain drug-disease intersection genes; the network of drug-active ingredients-disease-genes were constructed using Cytoscape 3.7.1 software; PPI of the intersection genes was built using the String online website; consequently to perform GO and KEGG of the intersection genes using R×64 3.5.3 software and Perl scripts. ResultsThere were six active ingredients of Haitongpi, 225 related targets and 14 intersecting genes between the drug and RA; the biological processes mainly involved in the treatment of RA with Haitongpi contained the activity of adrenergic receptor, the activity of G protein-coupled amine receptor, catecholamine binding and adrenergic receptor binding; the main signaling pathways involved covered cGMP-PKG signaling pathway, neuroactive ligand-receptor interaction signaling pathways and cCAMP. ConclusionHaitongpi could play the role in the treatment of RA through multiple components, multiple targets and multiple pathways, while the specific mechanism needs to be verified by zoopery and clinical trials.
