摘要:
目的探讨甘草甜素对病毒性脑炎小鼠神经元活性及血清神经元特异性烯醇化酶(neuronspecific enolase,NSE)的影响。 方法将SD小鼠30只随机分为生理盐水组、病毒性脑炎组和甘草甜素组,每组10只。注射柯萨奇病毒B3建立病毒性脑炎模型,甘草甜素组灌胃甘草甜素50 mg/kg,生理盐水组及病毒性脑炎组灌胃等剂量生理盐水。采用迷宫测试方法检测小鼠行为能力,甲酚紫(cresyl violet,CV)法检测神经元病理变化,酶联免疫法检测NSE与TUNEL法检测小鼠神经元凋亡情况,免疫印迹法检测神经元内Bax、Caspase-3。 结果3组小鼠逃避潜伏期与NSE值均随灌胃天数的增加而减少,其中病毒性脑炎组高于生理盐水组(P<0.05),甘草甜素组低于病毒性脑炎组(P<0.05);与生理盐水组相比,病毒性脑炎组S100B升高(P<0.05);与病毒性脑炎组相比,甘草甜素组S100B下降(P<0.05);小鼠神经元凋亡率,病毒性脑炎组高于生理盐水组(P<0.05),甘草甜素组低于病毒性脑炎组(P<0.05);生理盐水组小鼠几乎无Bax、Caspase-3蛋白表达,甘草甜素组Bax、Caspase-3蛋白相对表达量低于病毒性脑炎组(P<0.05)。 结论甘草甜素对病毒性脑炎小鼠行为能力及神经元活性改善有一定积极作用,其作用机制可能与调控NSE、S100B、Bax及Caspase-3水平有关。
Abstract:
ObjectiveTo explore the influence of glycyrrhizin on neuronal activity and serum NSE levels in mice suffering viral encephalitis. MethodsThirty mice were randomized into physiological saline group, viral encephalitis group and glycyrrhizin group, with ten in each group. Coxsackie virus B3 was injected to establish mice models with viral encephalitis, glycyrrhizin group accepted gastric lavage with glycyrrhizin at a dose of 50 mg/kg, physiological saline group and viral encephalitis group the equivalent dose of physiological saline. Maze test method was used to detect the behavioral ability of mice, cresyl violet (CV) to detect neuronal pathological changes, ELISA to measure NSE, TUNEL to detect neuron death in rats, Western bloting to detect the expressions of Bax and Caspase-3 in neurons. ResultsThe escape latency and NSE of the three groups decreased as the number of the gavage days increased, viral encephalitis group was higher than physiological saline group (P<0.05). Glycyrrhizin group was lower than viral encephalitis group (P<0.05); compared with physiological saline group, S100B of viral encephalitis group increased (P<0.05); compared with viral encephalitis group, S100B decreased in glycyrrhizin group (P<0.05); in terms of the mice's neuronal apoptosis rate, viral encephalitis group was higher than physiological saline group (P<0.05), glycyrrhizin group was lower than viral encephalitis group (P<0.05); there was almost no expression of BCL2-Associated X (BAX) and caspase-3 protein in the physiological saline group, the relative expressions of BAX and Caspase-3 protein in the glycyrrhizin group were lower than these of viral encephalitis group (P<0.05). ConclusionGlycyrrhizin has certain positive effects on the mice's behavioral ability and the activity of neurons, and its mechanism might be related to the regulation of the levels of NSE, S100B, Bax and Caspase-3.
