摘要:
目的探讨四逆汤通过自噬途径对脓毒症大鼠左心室功能的调控作用。 方法将150只雌性SD大鼠随机选取30只作为空白组(8 h、16 h、24 h各10只),其余120只按盲肠结扎穿刺法造模,将造模成功的90只大鼠按随机数字表方法分为实验组、对照组、阻断组各30只,每组再分为8、16、24 h组各10只。实验组灌胃四逆汤,对照组与空白组灌胃生理盐水,阻断组灌胃四逆汤+3-MA(6-氨基-3-甲基嘌呤)注射阻断,观察各组大鼠8、16、24 h心脏超声左心室收缩末期容积、左心室舒张末期容积、心输出量、左心室射血分数;并通过WB检测大鼠心肌自噬蛋白P62、LC3-I、LC3-Ⅱ水平。 结果1)干预8、16 h实验组大鼠左心室功能优于空白组、对照组、阻断阻(P<0.05),对照组与阻断组比较差异无统计学意义(P>0.05),空白组左心室功能优于对照组和阻断组(P<0.05);干预24 h大鼠左心室功能实验组、对照组、阻断组两两比较差异无统计学意义(P>0.05)。2)干预8、16 h实验组大鼠自噬蛋白表达强于对照组、阻断组(P<0.05),对照组与阻断组比较差异无统计学意义(P>0.05);干预24 h,大鼠自噬蛋白表达实验组、对照组、阻断组两两比较差异无统计学意义(P>0.05)。 结论四逆汤可能通过自噬途径改善脓毒症大鼠左心室功能。
Abstract:
ObjectiveTo discuss the regulatory effects of Sini Tang on left ventricular function in sepsis rats via autophagy. MethodsThirty rats were randomly chosen from 150 female SD rats as the blank group (ten rats at 8 h, 16 h and 24 h respectively), the remaining 120 were prepared into the models in light of cecal ligation puncture (CLP), 90 rats, successfully modelling, were divided into the experiment group, the control group and the blockade group, 30 cases in each group, and each group were subdivided into the groups of 8 h, 16 h and 24 h with ten in each. The experiment group was drenched with Sini Tang, the control group and the blank group with physiological saline, and the blockade group with Sini Tang plus 3-MA injection blockade, to observe LVEDV, LVESV, CO and LVEF of cardiac ultrasound in different groups at 8 h, 16 h and 24 h; and to detect the conditions of P62, LC3-? and LC3-Ⅱ of myocardial autophagy protein using WB. Results1)After intervention for 8 and 16 hours, left ventricular function of the experiment group was superior to that of the blank group, the control group and the blockade group (P<0.05), the difference had no statistical meaning in the control group and the blockade group (P>0.05); left ventricular function of the blank group was better than that of the control group and the blockade group (P<0.05); after intervention for 24 hours, the difference had no statistical meaning in left ventricular function between the experiment group, the control group and the blockade group (P>0.05). 2)After intervention for 8 and 16 hours, the expressions of autophagy protein of the experiment group were higher than these of the control group and the blockade group (P<0.05), the difference had no statistical meaning between the control group and the blockade group (P>0.05); after intervention for 24 hours, the difference had no statistical meaning in the expressions of autophagy protein between the experiment group, the control group and the blockade group (P>0.05). ConclusionSini Tang could improve sepsis rats' left ventricular function possibly through autophagy.
