摘要:
目的探讨木丹颗粒含药血清对高糖环境下雪旺细胞(schwann cells,SCs)自噬相关蛋白的调控作用。 方法将RSC96细胞分为25 mM葡萄糖组,150 mM葡萄糖组,150 mM葡萄糖+α-硫辛酸组和150 mM葡萄糖+木丹颗粒(10%,1%和0.1%)组,分别给予相应含药血清孵育48 h后,收集细胞用于RT-PCR法测定SCs中肿瘤坏死因子α(tumor necrosis factor,TNF-α)、白细胞介素6(interleukin-6,IL-6)、Beclin-1和微管相关蛋白1轻链3(microtubule-associated protein 1 light chain 3,LC3)的 mRNA表达,Western blot法测定TNF-α、IL-6、Beclin-1、LC3-I、LC3-II、磷酸化AMP激活的蛋白激酶(phosphorylation AMP-activated protein kinase,p-AMPK)及磷酸化哺乳动物雷帕霉素靶蛋白(phosphorylation mammalian target of rapamycin,p-mTOR)的蛋白表达。 结果与25 mM葡萄糖组比较,150 mM葡萄糖组的SCs活力显著降低(P<0.05),TNF-α、IL-6和p-mTOR的表达均显著升高(P<0.05),Beclin-1、LC-3和p-AMPK的表达,LC3-II/LC3-I比值均显著降低(P<0.05)。与150 mM葡萄糖组比较,150 mM葡萄糖+木丹颗粒组的SCs活力显著升高(P<0.05),TNF-α、IL-6和p-mTOR的表达均显著降低(P<0.05),Beclin-1、LC-3和p-AMPK的表达,LC3-II/LC3-I比值均显著升高(P<0.05)。 结论木丹颗粒含药血清能够通过增强AMPK的活性并抑制mTOR的活性,进而增强自噬活性,抑制炎症反应,从而改善高糖环境下的SCs损伤。
Abstract:
ObjectiveTo explore the regulatory effects of medicated serum of Mudan granules on autophagy-related proteins of SCs in high glucose environment. MethodsRSC 96 cells were divided into 25 mM glucose group, 150 mM glucose group, 150 mM glucose+α- Lipoic acid group, 150 mM glucose+Mudan granules (10%, 1% and 0.1%) group, after culturing for 48 hours with the corresponding serum containing the drugs respectively, the cells were collected to detect the expressions of LC3 mRNA, Beclin-1, IL-6 and TNF-α in SCs using RT-PCR method, Western blot method was used to measure the expressions of TNF-α, IL-6, Beclin-1, LC3-I, LC3-II, p-AMPK and p-mTOR protein. ResultsCompared with 25 mM glucose group, the activity of SCs reduced notably in 150 mM glucose group (P<0.05), the expressions of TNF-α, IL-6 and p-mTOR increased notably (P<0.05), the expressions of Beclin-1, LC-3 and p-AMPK, the ratio of LC3-II/LC3-I decreased signficantly (P<0.05); Compared with 150 mM glucose group, the activity of SCs raised notably in 150 mM glucose+Mudan granules group (P<0.05), the expressions of TNF-α, IL-6 and p-mTOR decreased notably (P<0.05), the expressions of Beclin-1, LC-3 and p-AMPK, the ratio of LC3-II/LC3-I increased signfi-cantly (P<0.05). ConclusionMedicated serum of Mudan granules could enhance the activity of autophagy and inhibit inflammatory reaction by boosting the activity of AMPK and restraining the activity of mTOR, therefore to improve SCs injury in high glucose environment.
