摘要:
目的运用网络药理学研究方法探讨真武汤合葶苈大枣泻肺汤治疗急性心力衰竭的作用机制。 方法在中药系统药理学分析平台上运用中药分子机制的生物信息学分析工具,筛选真武汤合葶苈大枣泻肺汤的成分和靶点;并通过相关数据库检索急性心力衰竭相关靶点;通过韦恩图得到真武汤合葶苈大枣泻肺汤和急性心力衰竭的共同靶点,然后利用STRING网站进行化合物-疾病共同靶点蛋白互作分析;利用David数据库进行基因本体分析和通路富集分析。最后,采用Cytoscape 3.72软件构建药物-成分-作用靶点-通路-疾病的网络模型。 结果通过筛选得到真武汤合葶苈大枣泻肺汤的潜在主要活性成分196个,涉及127个疾病作用靶点,主要活性成分为檞皮素、β-谷固醇、山柰酚、芍药苷等,信号通路为AGE-RAGE、IL-17等,共同发挥治疗急性心力衰竭的作用。 结论真武汤合葶苈大枣泻肺汤治疗急性心力衰竭是多成分、多靶点、多途径相互作用的结果,为真武汤合葶苈大枣泻肺汤的基础或临床研究提供一定的理论依据。
Abstract:
ObjectiveTo discuss the mechanism of Zhenwu decoction and Tingli Dazao Xiefei decoction in the treatment of acute heart failure (AHF) using network pharmacology. MethodsThe components and targets of Zhenwu decoction and Tingli Dazao Xiefei decoction were searched using the bioinformatics analysis tool of molecular mechanism of traditional Chinese medicine on TCMSP; AHF - related targets were retrieved through the relevant database; the targets shared by the decoction and AHF were gained by Venny diagram, compound-disease common target protein interaction analysis was carried out using STRING website; Gene ontology analysis and pathway enrichment analysis were conducted using David database. Consequently, Cytoscape 3.72 software was used to construct the drug-component-target-pathway-disease network model. ResultsBy searching, the study has obtained 196 potential main active ingredients of the decoction, including geranylgeranyl, β-glutathione, kaempferol, paeoniflorin, involving 127 targets of disease, and the related signal pathways were AGE-RAGE, IL-17 and others, playing the role of treating AHF jointly. ConclusionZhenwu decoction and Tingli Dazao Xiefei decoction in the treatment of AHF is the results of multi-ingredients, multi-targets and multi-pathways interacting on each other, and it could provide a certain theoretical reference for basic or clinical studies on the decoction.
