摘要:
目的:应用肾复康Ⅱ号联合间充质干细胞(MSCs)移植、肾复康Ⅱ号联合肝细胞生长因子拮抗剂(NK4)拮抗MSCs表面的肝细胞生长因子(HGF)后再移植的方法,研究肾复康Ⅱ号对MSCs的旁分泌作用,探讨其对MSCs旁分泌功能的作用机制。方法:本研究将50只雄性SD大鼠随机分为模型组、生理盐水+MSCs移植组、生理盐水+NK4拮抗HGF表达的MSCs组、肾复康Ⅱ号+MSCs移植组、肾复康Ⅱ号+NK4拮抗HGF表达的MSCs组,各组分别给予肾复康Ⅱ号405 mg/(kg·d)、等体积生理盐水灌胃,1次/d,连续14天。采用全骨髓贴壁筛选法体外分离、扩增培养大鼠MSCs,应用NK4蛋白转染质粒干预MSCs;单侧输尿管结扎的方法制备大鼠肾间质纤维化模型。通过尾静脉注射MSCs的方法进行移植。移植后4周对各组大鼠肾组织中HGF和特异性受体(C-Met)水平进行检测。结果:即时聚合酶链锁反应(RT-PCR)、蛋白质印迹法(Western blot)结果显示,肾复康Ⅱ号+MSCs组较其余各组肾组织HGF及c-Met均明显增高,肾复康Ⅱ号+MSCs组升高最明显,其次为肾复康Ⅱ号+拮抗剂组、生理盐水+MSCs组,各组之间比较,差异有统计学意义。结论:肾复康Ⅱ号具有促进MSCs在体内分泌HGF的作用,本研究验证了益肾散结方肾复康Ⅱ号对植入的MSCs具有旁分泌功能,并部分揭示了其调控机制。
Abstract:
Objective: To study the effects of ShenFuKang Ⅱ on paracrine function of mesenchymalstem cells (MSCs) and discuss its mechanism of ShenFuKang Ⅱ in promoting paracrine function of MSCs by using the method of ShenFuKang Ⅱ joined with MSCs transplantation, ShenFuKang Ⅱ joined to the retransplantation of MSCs after antagonizing superficial hepatocyte growth factor (HGF) by NK4. Methods: In the study, 50 male SD rats were randomized into the model group, the group of physiological saline+MSCs transplantation, the group of physiological saline+MSCs after antagonizing HGF expressions by NK4, the group of ShenFuKang Ⅱ+MSCs transplantation, and the group of ShenFuKang Ⅱ+MSCs after antagonizing HGF expressions by NK4, different groups were drenched by ShenFuKang Ⅱ, 405 mg/(kg·d) and physiological saline of equivalent volumes, once each day, for 14 days consecutively. Rat MSCs were isolated in vitro by whole marrow adherence screening method, cultured and proliferated, MSCs were intervened by NK4 protein transfecting plasmid; rat model suffering renal tubulointerstitial fibrosis was prepared by unilateral ureteral ligation. MSCs were transplanted by caudal vein injection. The levels of HGF and C-Met were detected in renal tissue of the rats in four weeks after transplantation. Results: The results of real-time quantitative PCR and Western blot demonstrated that the levels of HGF and C-Met in renal tissue of the group of ShenFuKang Ⅱ+MSCs were raised compared with the other groups obviously, the raise of the group of ShenFuKang Ⅱ+MSCs was the most obvious, the next was the group of ShenFuKang Ⅱ+antagonist, and the groups of physiological saline+MSCs, the differences were significant in the comparisons among different groups. Conclusion: ShenFuKang Ⅱ possesses the function of promoting MSCs secretinG HGF in vivo. The study tests and verifies that kidney-nourishing stasis-removing compound prescription ShenFuKang Ⅱ could induce paracrine action of the transplanted MSCs, and reveals its regulatory mechanism partly.
