摘要:
目的：研究涤痰通瘀方对急性脑出血大鼠肝组织核因子-κB(nuclear transcription factor-κB，NF-κB)的活性与肿瘤坏死因子α(tumor necrosis factor α，TNF-α)水平表达的干预作用。方法：将45只健康SD大鼠随机分为正常组、模型组及中药组，采用大脑注射VⅡ型胶原酶法建立大鼠急性脑出血模型，分别于造模后第24、48、72 h，采用EMSA检测大鼠肝组织NF-κB的活性，采用ELISA测定TNF-α的表达水平。结果：模型组大鼠的NF-κB活性明显高于正常组(P＜0.01)和中药组(P＜0.05)。ELISA法结果显示，与正常组TNF-α的浓度相比，模型组显著升高(P＜0.05)，而中药组明显降低(P＜0.05)。结论：中药组大鼠肝组织NF-κB及TNF-α的表达均较模型组低，说明涤痰通瘀方可以通过抑制LPS诱导活化核转录因子NF-κB，减少TNF-α炎性因子的合成释放，从而发挥对脑出血肝损害的保护作用。
Abstract:
Objective: To study the intervention effects of Ditan Tongyu prescription on the expressions of TNF-α and the activity of nuclear factor-κB in liver tissue of the rats with acute cerebral hemorrhage (ACH). Methods: All 45 healthy SD rats were randomized into the normal group, the model group and herbal group, intracerebral injection of VⅡ type collagenase were used to establish ACH rat models, EMSA method was adopted to detect the activity of NF-κB in liver tissue of the rats 24, 48 and 72 hours after modelling respectively, ELISA method was used to detect the expressions of TNF- α. Results: The activity of NF- κB of the rats in the model group was remarkably higher than that of the normal group (P＜0.01) and herbal group (P＜0.05). ELISA results showed that, compared with the concentrations of TNF- α in the normal group, it rose apparently in the model group (P＜0.05), while it lowered notably in herbal group (P＜0.05). Conclusion: The expressions of NF-κB and TNF-α in liver tissue of herbal group are lower than these of the model group, which demonstrates that Ditan Tongyu prescription could develop protective effects against liver damage in cerebral hemorrhage through inhibiting LPS, inducing the activation of NF- κB and reducing the synthesis and release of TNF-α.
