摘要:
目的采用网络药理学方法研究黄连抗焦虑的可能作用机制及靶点。 方法通过相关数据库获取黄连的主要活性成分及对应靶点,在疾病靶点数据库中获取焦虑症的靶点,筛选出两者相同靶点。通过Cytoscape软件构建“药物-成分-靶点-疾病”网络,采用STRING数据库构建蛋白互作网络(PPI网络),最后使用R软件及Bioconductor数据包进行GO富集分析(gene ontology,GO)和KEGG富集分析(kyoto encyclopedia of genes and genomes,KEGG)。 结果共筛选到黄连活性成分14种和对应作用靶点84个,包括白细胞介素6(interleukin-6,IL-6)、胱天蛋白酶3(Caspase-3,CASP3)、雌激素受体1(Estrogen receptor 1,ESR1)、禽髓细胞瘤病毒癌基因同源物(V-Myc avian myelocytomatosis viral oncogene homolog,MYC)、雄激素受体(androgen receptor,AR)等。GO富集分析得到生物过程条目721个,细胞组分条目36个、分子功能条目83个;KEGG通路富集分析得到78条信号通路,主要干预糖尿病并发症的AGE-RAGE信号通路、流体剪切应力与动脉粥样硬化、卡波西肉瘤相关疱疹病毒感染等信号通路。 结论黄连主要通过参与调节脑内神经递质、抗炎、抗氧化应激、调节内分泌及免疫紊乱等发挥抗焦虑作用,是一个多成分、多靶点、多通路的复杂干预过程。
Abstract:
ObjectiveTo study the possible mechanism and targets of Huanglian (Coptidis rhizoma) fighting anxiety based on network pharmacology method. MethodsThe main active ingredients and the corresponding targets of Huanglian were obtained via the relevant database, the targets of anxiety were gained from disease target database, to find out the common targets of anxiety and Huanglian. Cytoscape software was used to construct the network of "medicine-ingredient-target-disease", STRING database was adopted to establish PPI, finally, R software and Bioconductor data package were utilized to perform GO and KEGG. ResultsAll 14 active ingredients from Huanglian and 84 corresponding targets were screened out, including IL-6, CASP3, ESR1, MYC, AR and others. All 721 biological process items, 36 cell component items and 83 molecular function items were gained by GO enrichment analysis; 78 signaling pathways were obtained by KEGG pathway enrichment analysis, which mainly interferes with AGE-RAGE signaling pathway, fluid shear stress and atherosclerosis, Kaposi's sarcoma associated herpes virus infection in diabetic complications. ConclusionHuanglian has an anti-anxiety effects mainly through participating in the regulation of brain endogenous transmitters, anti-inflammatory, antioxidant stress, endocrine regulation and immune disorders, which is a complex intervention involving many ingredients, many targets and pathways.
