摘要:
目的探讨生姜治疗阿尔茨海默病(Alzheimer's disease,AD)的潜在主要活性成分、作用机制。 方法使用中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP),以“生姜”为检索词,获取生姜活性成分;以AD为关键词,获取OMIM、GeneCards等数据库中AD靶点。取药物治疗AD的核心靶点,将核心靶点导入String库,绘制蛋白互作网络,并筛选出蛋白核心靶点。针对靶点进行GO和KEGG富集分析。 结果检索到生姜主要活性成分5个,共同靶标60个,主要活性成分分别为beta-sitosterol、6-methylgingediacetate2、Stigmasterol、poriferast-5-en-3beta-ol、Dihydrocapsaicin等;基因靶点以IL-6、AKT1、JUN、MAPK8、GASP3、PTGS2、CAT等为主,主要筛选出5条信号通路,包括AGE/RAGE、TNF、IL-17、C-type lectin receptor、Toll样受体等通路。 结论生姜可能通过抗炎、抗氧化、促进神经再生等多层次、多途径、多靶点发挥对AD的治疗作用。
Abstract:
ObjectiveTo explore the mechanism and the potential main active ingredients of ginger in the treatment of Alzheimer's disease (AD). MethodsUsing TCMSP, "ginger" was taken as the term, to obtain the active ingredients of ginger; AD was chosen as the term to gain AD targets from OMIM and GeneCards databases. The core targets of drug therapy for AD were chosen and imported into String database to draw PPI network and screen out the core targets of the protein. GO and KEGG enrichment analysis were performed for the targets. ResultsWe have obtained five main active ingredients of ginger, 60 common targets, and the main active ingredients were beta-sitosterol, 6-methylgingediacetate2, stigmasterol, poriferast-5-en-3beta-ol and dihydrocap- saicin respectively; genetic targets contained IL-6, AKT1, JUN, MAPK8, GASP3, PTGS2, CAT, etc, and five signaling pathways were selected, covering AGE/RAGE, TNF, IL-17, C-type lectin receptor and Toll-like receptors. ConclusionGinger could play the therapeutic role in the treatment of AD possibly through anti-inflammatory, anti-oxidation and promoting nerve regeneration from multi-level, multi-pathway and multi-targets.
