摘要:
目的探究清热解毒法与益气养阴法代表药对治疗放射性口腔黏膜炎(radiation-induced oral mucositis,RIOM)的作用靶点及机制。 方法通过文献检索获得代表药对,利用中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)与中药分子机制生物信息学分析工具(bioinformatics analysis tool for molecular mechanism of traditional Chinese medicine,BATMAN-TCM)数据库获取药物成分及靶点,从Drugbank数据库、GeneCards数据库、人类孟德尔遗传在线数据库(online mendelian inheritance in man,OMIM)获取疾病靶点。构建蛋白质-蛋白质相互作用(protein-protein interaction,PPI)网络和“药对-化合物-通路-疾病”网络,进行京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)富集分析,采用AutoDock Vina软件进行分子对接。 结果清热解毒法代表药对为金银花-连翘,益气养阴法代表药对为北沙参-麦冬。两药对共有靶点104个,涉及蛋白激酶B1(AKT1)、肿瘤蛋白p53(TP53)、转录因子Jun(JUN)、血管内皮生长因子A(vascular endothelial growth factor A,VEGFA)等;清热解毒法特有靶点30个,涉及白蛋白(albumin,ALB)、连环蛋白β1(catenin beta-1,CTNNB1)、小鼠双微体2同源基因(mouse double minute 2 homolog,MDM2)等;益气养阴法特有靶点39个,涉及乳腺癌易感基因1(breast cancer susceptibility gene 1,BRCA1)、过氧化氢酶(catalase,CAT)、端粒酶逆转录酶(telomerase reverse transcriptase,TERT)等。共同靶点KEGG通路187条,涉及癌症信号通路、肿瘤坏死因子信号通路、白细胞介素17信号通路等;清热解毒法特有通路91条,涉及细胞周期通路、p53信号通路等;益气养阴法特有通路12条,涉及酪氨酸代谢通路、嘧啶代谢通路等。分子对接显示关键靶点与活性成分结合稳定。 结论两治法治疗RIOM存在共同靶点与通路(癌症信号通路、细胞凋亡通路等),亦各有特点(清热解毒法通过神经活性配体-受体相互作用,益气养阴法通过嘧啶代谢信号通路等),为综合运用及针对性治疗提供了理论依据。
Abstract:
ObjectiveTo explore the targets and mechanisms of representative herb pairs of heat-clearing and detoxifying method and Qi-replenishing and Yin-nourishing method in the treatment of radiation-induced oral mucositis (RIOM). MethodsRepresentative herb pairs were obtained through literature search. The active components and corresponding targets of the herbs were retrieved from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) and the Bioinformatics Analysis Tool for Molecular Mechanism of Traditional Chinese Medicine (BATMAN-TCM) database. Disease targets were obtained from the DrugBank database, GeneCards database, and Online Mendelian Inheritance in Man (OMIM). A protein-protein interaction (PPI) network and a "herb pair-compound-pathway-disease" network were constructed, followed by Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. Molecular docking was performed using AutoDock Vina software. ResultsThe representative herb pair of heat-clearing and detoxifying method was Jinyinhua (Lonicerae Japonicae Flos)-Lianqiao(Forsythiae Fructus), and that of Qi-replenishing and Yin-nourishing method method was Beishashen ( Glehniae Radix)-Maidong (Ophiopogonis Radix). A total of 104 common targets were identified for the two herb pairs, involving AKT serine/threonine kinase 1 (AKT1), tumor protein p53 (TP53), Jun proto-oncogene (JUN), vascular endothelial growth factor A (VEGFA), etc. There were 30 unique targets of heat-clearing and detoxifying method, involving albumin (ALB), catenin beta-1 (CTNNB1), mouse double minute 2 homolog (MDM2), etc., and 39 unique targets of Qi-replenishing and Yin-nourishing method, involving breast cancer susceptibility gene 1 (BRCA1), catalase (CAT), telomerase reverse transcriptase (TERT), etc. A total of 187 KEGG pathways were shared by the common targets, involving the pathways in cancer, tumor necrosis factor (TNF) signaling pathway, interleukin-17 (IL-17) signaling pathway, etc. There were 91 unique pathways of heat-clearing and detoxifying method, involving the cell cycle pathway, p53 signaling pathway, etc., and 12 unique pathways of Qi-replenishing and Yin-nourishing method, involving tyrosine metabolism and pyrimidine metabolism pathways. Molecular docking results demonstrated stable binding between the key targets and the active components. ConclusionThe two therapeutic methods share common targets and pathways (such as the pathways in cancer and apoptosis signaling pathway) in the treatment of RIOM, while each also possesses distinct characteristics, heat-clearing and detoxifying method primarily acts through the neuroactive ligand-receptor interaction pathway, and Qi-replenishing and Yin-nourishing method mainly exerts its effects via the pyrimidine metabolism signaling pathway, among others. These findings provide a theoretical basis for the integrated application and targeted treatment of RIOM.
