摘要:
目的:明确昆仑雪菊中原花青素B2和绿原酸的含量,探讨昆仑雪菊对高脂饮食2型糖尿病小鼠血糖的影响。方法:采用RP-HPLC法,以流动相乙腈-0.01%磷酸溶液,检测波长315 nm,流速1 mL/min测定昆仑雪菊中原花青素B2和绿原酸的含量。将昆明种小鼠60只随机分为对照组、模型组、阳性组及昆仑雪菊高(4 g/kg)、中(2 g/kg)、低(1 g/kg)剂量组,对照组与模型组给予等容量生理盐水,其余各组灌胃给予相应药物,连续14天。在造模前后以及给药后7天和14天测定各组小鼠血糖值,并观察小鼠体质量和超氧化物歧化酶(SOD)活力及丙二醛(MDA)含量。结果:昆仑雪菊中原花青素B2和绿原酸的含量分别为0.016、1.683 mg/g。昆仑雪菊能明显升高糖尿病模型小鼠体质量(P<0.05),降低糖尿病模型小鼠血糖(P<0.05),升高糖尿病模型小鼠SOD的活力,降低MDA含量。结论:昆仑雪菊对高脂饮食2型糖尿病小鼠有一定保护作用,其有效成分可能是原花青素B2和绿原酸。
Abstract:
Objective: To detect the contents of procyanidine B2 and chlorogenic acid contained in KunLun chrysanthemum, and discuss the effects of Kunlun chrysanthemum on the levels of blood glucose of the mice with type two diabetes mellitus(T2DM) prepared by high fat diet. Methods: The contents of procyanidine B2 and chlorogenic acid contained in Kunlun chrysanthemum were detected by using RP-HPLC method with the mobile phase of acetonitrile-0.01% phosphoric acid solution. The detection wavelengthen was 315nm, flow rate was 1 mL/min. Sixty mice were randomized into the control group, the model group, the positive group, high(4 g/kg), moderate(2 g/kg) and low(1 g/kg) doses groups of Kunlun chrysanthemum, the control group and the model group accepted physiological saline in the same volumes, other groups were drenched with the corresponding medicine, for 14 days consecutively. The values of blood glucose of the mice in different groups were measured before and after modeling, in seven days and 14 days after medication, the contents of MDA, the activity of SOD and body weight of the mice were observed. Results: The contents of procyanidine B2 and chlorogenic acid contained in Kunlun chrysanthemum were 0.016 mg/g and 1.683mg/g respectively. Kunlun chrysanthemum could obviously raise body weight of diabetic mice(P<0.05), lower the blood glucose of diabetic mice (P<0.05), improve the activity of SOD and decrease the contents of MDA. Conclusion: KunLun chrysanthemum shows certain protective effects to the mice with T2DM, and its active ingredients might be procyanidine B2 and chlorogenic acid.
