摘要:
目的:研究扶正祛邪双表法对流感病毒感染小鼠肺组织TLR4/NF-κB信号通路的调控作用。方法:将昆明种小鼠随机分为模型组、正常组、解表法代表方剂银翘散(YQS)组、固表法代表方剂玉屏风散(YPF)组、双表法代表方剂(YPS)组以及利巴韦林组。采用流感病毒滴鼻方法建立病毒感染模型,根据给药量制备相应浓度药物并灌胃给药,正常组和模型组给予同体积生理盐水。采用RT-PCR、免疫组化法检测给药后1、3、5、7天各时间点各组小鼠Toll样受体4(toll like receptors 4,TLR4)和核转录因子κB(nuclear factor-κB,NF-κB)蛋白及其mRNA动态表达情况。结果:YQS、YPF及YPS在不同时点均能不同程度阻断流感小鼠肺组织中TLR4、NF-κB蛋白及其mRNA的高表达,并且YPS组表达最低,差异有统计学意义(P<0.05)。结论:双表法对流感病毒感染小鼠的防治作用与抑制流感病毒感染小鼠肺组织TLR4/NF-κB信号通路过度活化有关。
Abstract:
Objective: To study the regulatory effects of double exterior method of strengthening the body resistance and eliminating pathogenic factors on TLR4/NF-κB signal path of lung tissue in influenza virus-infected mice. Methods: Kunming mice were randomized into the model group, the normal group, the group of Yinqiao San (YQS) (the representative prescription of exterior-relieving method), the group of Yupingfeng San (YPF) (the representative prescription of exterior-consolidating method), the group of the representative prescription of double exterior method (YPS) and ribavirin group. Nose dropping method of influenza virus was used to establish virus-infection model, the concentrations of the drugs were prepared according to the dose and given to the models, the normal group and the model group were drenched with equivalent volume of normal saline. RT-PCR and immunohistochemical method were adopted to detect the dynamic expressions of mRNA, NF-κB protein and TLR4 of the mice in different groups at different time points one day, three, five and seven days after the medication. Results: YQS, YPF and YPS could block high expressions of mRNA, NF-κB protein and TLR4 in lung tissue of the mice model at different time points to different degrees, and the expressions of YPS group the lowest, and the difference had statistical meaning (P<0.05). Conclusion: Preventive and therapeutic effects of double exterior method for influenza virus-infected mice is related to inhibiting the overactivation of lung TLR4/NF-κB signal path of the mice model.
