摘要:
目的:观察桂枝茯苓胶囊对卵巢早衰(premature ovarian failure,POF)大鼠性激素水平、卵巢细胞凋亡的改善作用,并探讨其作用机制。方法:取50只健康雌性大鼠,其中40只采用腹腔注射环磷酰胺建立POF大鼠模型,将其随机分为POF组,桂枝茯苓胶囊低、中、高剂量组,剩余10只腹腔注射无菌生理盐水,设为对照组。桂枝茯苓胶囊低、中、高剂量组大鼠分别灌胃0.6、1.2、2.4 g/kg桂枝茯苓胶囊悬浮液2 mL,每日1次,POF组和对照组灌胃生理盐水2 mL,每日1次,连续4周。对比各组血清卵泡刺激激素(follicle stimulating hormone,FSH)、促黄体生成素(luteinizing hormone,LH)及雌二醇(estrogen,E2)水平;观察卵巢组织病理学改变,并对比各组卵巢细胞凋亡情况;对比卵巢组织中丝裂原活化蛋白激酶(p-38 mitogen-activated protein kinase,p-38 MAPK)、凋亡相关因子(factor associated suicide,Fas)、Fas配体(factor associated suicide ligand,FasL)、天冬氨酸特异性半胱氨酸蛋白酶3(Caspase-3)mRNA和蛋白表达情况,对比p-p38 MAPK/p38 MAPK蛋白比值。结果:血清FSH、LH由低到高、E2由高到低依次如下:对照组、桂枝茯苓胶囊高、中、低剂量组,POF组,每2组间比较差异均有统计学意义(P<0.05);卵巢组织病理学观察,对照组卵巢体积、各级卵泡细胞、黄体发育正常;POF组卵巢萎缩、卵泡、黄体少,闭锁卵泡增多;桂枝茯苓胶囊组各级卵泡细胞生长情况较POF组改善,其中高剂量组有明显改善;卵巢细胞凋亡率、卵巢组织中Fas、FasL、Caspase-3 mRNA和蛋白相对表达量、p-p38 MAPK/p-38 MAPK蛋白比值由低到高依次为对照组、高、中、低剂量组,POF组,每两组间比较差异均有统计学意义(P<0.05);p-38 MAPK mRNA相对表达量比较,差异无统计学意义(P>0.05)。结论:桂枝茯苓胶囊可显著改善POF大鼠性激素水平、减少卵巢细胞凋亡,其中中剂量组效果最佳,可能与抑制Fas/FasL信号通路有关。
Abstract:
Objective: To survey the effects of Guizhi Fuling capsules on improving the levels of sex hormone and the apoptosis of ovarian cells in the rats with premature ovarian failure (POF), and explore its mechanism. Methods: All 50 healthy female rats were chosen, among them, 40 ones were prepared into POF rat models using intraperitoneal injection of cyclophosphamide, and divided into POF group, low, moderate and high doses groups randomly, the remaining ten rats accepted intraperitoneal injection of sterile normal saline and they were set as the control group. Low, moderate and high doses groups were drenched with Guizhi Fuling capsules respectively in the concentrations of 0.6, 1.2 and 2.4 g/kg, POF group and the control group with normal saline, 2 mL, once each day, for four weeks consecutively. To investigate the levels of FSH, LH and E2 between different groups; to observe histopathological changes of ovary, compare the apoptosis of ovarian cells in different groups; to survey the expressions of p-38 MAPK, Fas, FasL, Caspase-3mRNA and protein in ovarian tissue, and explore p-38 MAPK/p38MAPK protein ratio. Results: FSH and LH from low to high, and E2 from high to low were as follows: the control group, high dose group, moderate dose group, low dose group and POF group, the differences were significant between every two groups (P<0.05); histopathological observation of ovary, ovarian volume, different levels of ovarian cells and corpus luteum developed normally in the control group; ovarian shrank, ovarian follicle amd corpus luteum lessened, atresia follicle increased in POF group; the growth of follicle cells at different levels in low, moderate and high doses groups were improved compared with these of POF group, among them, it was improved notably in high dose group; apoptosis rate of ovarian cells, the relative expressions of Fas, FasL, Caspase-3mRNA and protein in ovarian tissue, the ratio of p-38 MAPK/p38MAPK ranked from low to high as follows: the control group, high dose group, moderate dose dose group, low dose group and POF group, the differences were significant between every two groups (P<0.05); the difference was insignificant in the relative expression of p-38 MAPK mRNA (P>0.05). Conclusion: Guizhi Fuling capsules could significantly improve the levels of sex hormone in POF rats, reduce the apoptosis of ovarian cells, the effects are the best in the concentration of 1.2 g/kg, and it might be related to inhibiting Fas/FasL signaling pathway.
