摘要:
目的探讨腹部推拿对慢传输型便秘(slow transit constipation,STC)大鼠体内神经递质及5-羟色胺(5-hydroxytryptamine,5-HT)受体表达的作用机制。 方法将39只雄性大鼠随机分为对照组、模型组及推拿组,每组13只。模型组及推拿组大鼠采用肠神经节消融术建立STC大鼠模型,对照组不做处理,相同条件下继续喂养,造模成功后按照特定手法对推拿组STC大鼠进行连续14天的腹部推拿,对照组及模型组只摆相同体位,但不进行推拿;收集各组大鼠粪便,计算粪便含水率,测定大鼠血浆中P物质(substance P,SP)、一氧化氮合酶(nitric oxide synthase,NOS)及血管活性肠肽(vasoactive intestinal peptide,VIP)水平,测定大鼠首粒黑便排出时间及肠推动率,采用HE染色法进行大鼠结肠组织病理学检查,Western blot检测大鼠结肠组织5-HT3R、5-HT4R水平。 结果与对照组相比,模型组STC大鼠粪便含水率、血浆SP水平及结肠组织5-HT3R、5-HT4R水平均下降(P<0.05),血浆VIP及NOS水平升高(P<0.05),首粒黑便排出时间延长(P<0.05),肠推动率降低(P<0.05);HE染色结果表明模型组大鼠结肠组织发生病理性改变。与模型组相比,推拿组STC大鼠粪便含水率、血浆SP水平及结肠组织5-HT3R、5-HT4R水平均升高(P<0.05),血浆VIP及NOS水平降低(P<0.05),首粒黑便排出时间缩短(P<0.05),肠推动率升高(P<0.05),HE染色结果表明推拿组STC大鼠结肠组织病理学改变改善,细胞排列整齐,形态圆润。 结论腹部推拿能够改善STC大鼠疾病状态,调节大鼠体内神经递质及5-HT受体表达。
Abstract:
ObjectiveTo explore the mechanism of abdominal massage on the expression of neurotransmitters and 5-HT receptors in rats with slow transit constipation (STC). MethodsAll 39 male rats were randomly divided into three groups: the control group, the model group and the massage group, 13 rats in each group. The rats in the model group and the massage group were prepared into rat model with STC by intestinal ganglion ablation, the control group was unhandled, and the rats were fed under the same conditions continuously. After successful modeling, STC rats in the massage group were given abdominal massage for 14 days according to the specific technique, the rats in the control group and the model group were only in the same position,but not massage; To calculate the water content of feces after collecting the feces of rats in each group, the levels of substance P (SP), nitric oxide synthase (NOS) and vasoactive intestinal peptide (VIP) in plasma were measured, and the excretion time of the first black stool and intestinal propulsion rate were detected. The histopathological examination of rat colon was performed by HE staining, and the levels of 5-HT3R and 5-HT4R in rat colon tissue were detected by Western blot. ResultsCompared with the control group, the fecal water content, the levels of plasma SP and colonic 5-HT3R, 5-HT4R of STC rats in the model group decreased (P<0.05), the levels of plasma VIP and NOS rose (P<0.05), discharge time of first black stool prolonged (P<0.05), and the intestinal motility decreased significantly (P<0.05); HE staining results showed that pathological changes occurred in the colon tissue of rats in the model group. Compared with the model group, the fecal water content, the levels of plasma SP and colonic 5-HT3R, 5-HT4R of STC rats in the massage group raised (P<0.05), the levels of plasma VIP and NOS reduced (P<0.05), discharge time of first black stool shortened (P<0.05), and the intestinal motility rose significantly (P<0.05), the results of HE staining demonstrated that pathological changes of colon tissue in STC rats of the massage group improved, the cells were well arranged and round in shape in the massage group. ConclusionAbdominal massage could improve the disease status and regulate the expression of neurotransmitters and 5-HT receptors in STC rats.
