摘要:
目的基于网络药理学方法探讨八珍汤促宫颈成熟的分子作用机制和生物通路。 方法通过中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)获取八珍汤药物的有效化学成分并预测有效化学成分对应的靶点和基因,通过人类基因数据库(GeneCards)、孟德尔人类遗传数据库(online Mendelian inheritance in man,OMIM)、DiGSeE数据库、Drug Bank数据库和治疗靶标数据库(therapeutic target database,TTD)搜索得到宫颈成熟相关的疾病靶点基因,将药物靶点和疾病靶点进行比对,获得八珍汤促宫颈成熟的作用靶点并绘制韦恩(Venn)图。通过STRING官网(https://string-db.org/)构建蛋白互作(protein protein interaction,PPI)网络,并运用软件Cytoscape 3.7.2构建“促宫颈成熟-基因-靶点-八珍汤有效成分”的调控网络,筛选出核心基因。最后进行基因本体(gene ontology,GO)功能富集分析、京都基因与基因组百科全书(ryoto encyclopedia of genes and genomes,KEGG)通路富集分析其生物过程、细胞成分、分子功能和生物作用通路。 结果共筛选出160个化学成分,175个与促宫颈成熟相关的作用靶点,β-谷固醇、芒柄花黄素、谷甾醇、山柰酚、原阿片碱等为关键药物化学成分,前列腺素过氧化物合酶2(prostaglandin-endoperoxide synthase 2,PTGS2)、雌激素受体(estrogen receptor,ESR1)、雄激素受体(androgen receptor,AR)、肾上腺素能受体2(adrenrgic receptor 2,ADRB2)、核受体辅激活蛋白2(nuclear receptor coactivator 2,NCOA2)等为关键靶标。经过GO功能富集分析和KEGG通路富集分析,发现八珍汤通过参与了雌激素、催产素、松弛素信号通路等内分泌激素信号通路和白细胞介素17(interleukin-17,IL-17)信号通路、白细胞跨内皮转移等炎症反应信号通路促宫颈成熟。 结论八珍汤促宫颈成熟的有效成分、靶标和生物通路体现了中药多成分-多靶点-多途径的特点,与中医的“整体观念”一致,可为阐释八珍汤促宫颈成熟的基础研究和药物研究提供依据和参考;其关键化学成分和靶标揭示八珍汤中关键药物促宫颈成熟的作用可为八珍汤促宫颈成熟的用药用量规律基础研究和临床研究提供参考。
Abstract:
ObjectiveTo explore molecular mechanism and biologial pathway of Bazhen decoction in promoting cervical maturation based on network pharmacological method. MethodsThe active chemical components of Bazhen decoction were obtained by TCMSP, and the corresponding targets and genes of the active chemical components were predicted, target genes of cervical maturation related disease were searched by GeneCards, OMIM, DiGSeE, Drug Bank and TTD databases, the targets of Bazhen decoction in promoting cervical maturation were gained by comparing the drug targets with disease targets, to draw Venn diagram. PPI net was constructed by STRING official website, Cytoscape3.7.2 was used to construct the regulation network of “promoting cervical maturation-gene-target-active ingredients of Bazhen decoction”, and screen the core genes. Ultimately, GO and KEGG pathways were performed to analyze the biological process, cellular ingredients, molecular functions and biological pathways. ResultsAll 160 kinds of chemical ingredients were selected from Bazhen decoction, 175 targets related to promoting cervical maturation, β-sitosterol, formononetin, sitosterol, kaempferol and fumarine are the key pharmaceutical chemical components, PTGS2, ESR1, AR, ADRB2 and NCOA2 are the key targets. It is found that Bazhen decoction promotes cervical maturation by participating in endocrine hormone signaling pathways such as estrogen, oxytocin, relaxin signaling pathway, IL-17 signaling pathway, leukocyte transendothelial metastasis and other inflammatory signaling pathways via GO and KEGG pathway enrichment analysis. ConclusionThe active ingredients, targets and biological pathways of Bazhen decoction promoting cervical maturation embody the characteristics of multi-component, multi-target and multi-channel of traditional Chinese medicine, they are consistent with the “holism concept” of traditional Chinese medicine, it provides the basis and reference for explaining the basic research and drug research of Bazhen decoction promoting cervical maturation; its key chemical components and targets reveal the role of key drugs in Bazhen decoction in promoting cervical maturation, which can provide reference for the basic research and clinical research of the dosage law of Bazhen decoction in promoting cervical maturation.
