摘要:
目的建立吴茱萸UPLC指纹图谱分析方法,对不同产地吴茱萸药材质量进行全面评价,为控制左金方中吴茱萸药材质量提供依据。 方法安捷伦Infinity 1290 Ⅱ高效液相色谱仪;Ultimate UHPLC AQ-C18色谱柱(2.1×100 mm,1.8 ?m),以乙腈(A)-0.01%三氟乙酸(B)梯度洗脱,流速为每分钟0.4 mL;检测波长为300 nm,进样量1 μL。采收集了17批不同产地吴茱萸样品进行指纹图谱分析,结合主成分分析及有效成分含量测定,对吴茱萸药材整体质量进行评价。 结果建立了吴茱萸药材UPLC指纹图谱,标定了14个共有峰。指纹图谱和主成分分析显示不同产地吴茱萸药材指纹图谱基本一致,化学成分种类基本相同,成分的相对含量在一定范围内波动。吴茱萸药材质量与果实成熟度有较大的相关性,成熟度较低的样品柠檬苦素含量较高,吴茱萸碱和吴茱萸次碱总量较低,而成熟度较高药材与之相反。 结论指纹图谱结合有效成分含量测定的方法可用于吴茱萸药材的整体质量评价,为左金方中吴茱萸药材质量控制提供依据。
Abstract:
ObjectiveTo provide the reference for quality control of Wuzhuyu (Euodiae fructus) in Zuojin prescription by establishing the method of analyzing UPLC fingerprint of Wuzhuyu and comprehensively assess the quality of Wuzhuyu from different producing areas. MethodsThe conditions for chromatography were Ultimate UHPLC AQ-C18 column (2.1 × 100 mm, 1.8 μm) with Agilent Infinity 1290 II HPLC, acetonitrile and 0.01% trifluoroacetic acid (TFA) solution as the mobile phase was used to perform gradient elution, with the velocity of 0.4 mL?min-1; detection wavelength was 300 nm, and the sample size was 1 μL. The overall quality of Wuzhuyu was assessed by fingerprint analysis of 17 batches of samples from different producing areas, combined with principal component analysis (PCA), and the content determination of the active ingredients. ResultsAll 14 common peaks were calibrated in UPLC fingerprint of Wuzhuyu. Fingerprint and PCA revealed that Wuzhuyu from different producing areas had the basically identical fingerprints, and the basically same sorts of chemical ingredients, while the relative content of the components fluctuated within a certain range. The quality of Wuzhuyu has a great correlation with the maturity of the fruit, the samples with lower maturity had a higher content of limonin, and a lower total amount of evodiamine and rutecarpine, whereas the opposite was true for higher maturity samples. ConclusionFingerprint in combination with the content determination of the active ingredients could be used for the evaluation of overall quality of Wuzhuyu, and it could provide the reference for quality control of Wuzhuyu in Zuojin prescription.
