摘要:
根据心血管疾病(cardiovascular disease,CVD)血管钙化特点及机制,就缺氧诱导因子1α(hypoxia inducible factor 1 alpha,HIF-1α)与慢性肾脏病(chronic kidney disease,CKD)患者血管钙化的密切关系进行综述,CVD是CKD首要致死并发症,约占CKD患者总死亡率的50%;血管钙化是CVD发生的首要原因,与肾功能进展呈正相关;CKD患者的低氧状态是促进血管平滑肌细胞成骨样转分化及钙化的重要因素,而HIF-1α是细胞缺氧反应的关键转录调节因子,不仅可以通过介导多种成骨标志物,促进血管平滑肌细胞成骨样分化,参与血管钙化形成,还可通过诱导炎症及氧化应激、参与糖酵解途径、诱导细胞凋亡和细胞自噬等途径参与血管钙化的发生发展。
Abstract:
On the foundation of the characteristics and mechanism of vascular calcification in cardiovascular disease (CVD), the paper summed up the close relationship between HIF-1α and vascular calcification (VC) in CVD patients, CVD is the leading fatal complication of chronic kidney disease (CKD), approximately occupying 50% of total mortality rate in CKD patients. VC as the primary cause of CVD is positively correlated with the progress of renal function. Hypoxia is an important factor in promoting osteoblast-like transdifferentiation and calcification of vascular smooth muscle cells (VSMCs) in CKD patients, however, hypoxia-inducible factor-1α (HIF-1α) is a key transcriptional regulatory factor in cell hypoxia response, it could not only promote osteoblast-like differentiation of VSMCs and participate in the formation of VC by mediating a variety of osteogenic markers, but also take part in the attack and development of VC through inducing inflammation and oxidative stress, participating in glycolytic pathway as well as inducing cellular apoptosis and autophagy.
