摘要:
目的：探讨心脑通配方对大鼠颈动脉粥样硬化斑块中基质金属蛋白酶-2(MMP-2)及血脂的影响。方法：将50只SD大鼠随机分为5组,每组10只，分别给予正常饲料喂养、左侧颈总动脉血管钳夹术加高脂饮食喂养及血管钳夹术加分别含有低、中、高3个剂量浓度心脑通配方的高脂饲料喂养。于3周、8周、12周测血脂水平；采用免疫组织化学染色法观察MMP-2在颈动脉中的表达和分布。结果：①与模型组相比,心脑通3个剂量组能够降低大鼠血清TC、TG、LDL-C水平(P＜0.05)，提高HDL-C(P＜0.05)；②MMP-2在正常对照组大鼠颈总动脉组织内低水平表达，在钳夹高脂饲养组大鼠颈总动脉组织中表达水平明显升高(P＜0.05)，且明显高于正常对照组（P＜0.01）。而心脑通3个剂量组中MMP-2含量低剂量组>中剂量组>高剂量。结论：①动脉粥样硬化模型大鼠颈总动脉组织粥样硬化斑块中有MMP-2表达；②心脑通配方可能通过抑制MMP-2表达而抑制斑块增殖和破溃，从而抑制动脉粥样硬化斑块的形成。
Abstract:
Objective: To explore the effects of XinNaoTong prescriptions on MMP-2 and blood lipid of the plaque in rats with carotid atherosclerosis. Methods: All 50 SD rats were randomized into five groups, ten rats each group, all the rats were fed with normal diets, left carotid artery clamped and fed with high cholesterol diet, artery clamped and high cholesterol diet with low, moderate and high dosages of XinNaoTong prescription respectively. The levels of blood lipid were detected at the 3rd, 8th and 12th week; the expressions and the distribution of MMP-2 in the carotid artery were observed by immunohistochemical staining method. Results: ① compared with the model group, three groups of XinNaoTong could lower the levels of TC, TG and LDL-C in the serum(P＜0.05); raise the level of HDL-C (P＜0.05); ② the levels of MMP-2 were lower in carotid artery of normal control group, it was raised in the tissue of carotid artery of the group with clamped and high cholesterol diet (P＜0.05), and obviously higher than normal control group(P＜0.01). The orders of MMP-2 contents in XinNaoTong groups were: low dosage group＞moderate dosage group＞high dosage group. Conclusion: ① there are MMP-2 expressions in the plaque of rats with carotid atherosclerosis. ② XinNaoTong could inhibit the proliferation and ulceration of the plaque by inhibiting MMP-2 expressions, therefore to hinder the formation of atherosclerosis plaque.
