摘要:
目的：探讨温肾活血化湿方对单侧输尿管梗阻(unilateral ureteral obstruction，UUO)法致肾间质纤维化大鼠肾组织核转录因子-κB(NF-κB)表达的影响，初步揭示该方防治肾间质纤维化的作用机制。方法：采用UUO建立大鼠肾间质纤维化模型，将雄性SD大鼠80只随机分为假手术组、UUO组、氯沙坦组和温肾活血化湿方组，每组20只；于造模后第2天开始灌胃，假手术组、UUO组给予等量生理盐水，氯沙坦组灌胃氯沙坦［10 mg/(kg·d)］，温肾活血化湿方组灌温肾活血化湿方汤剂［9.8 mg/(kg·d)］。于造模后第7、14、21天留取梗阻侧肾组织行HE、Masson染色，观察肾组织病理变化，免疫组化法检测肾组织NF-κB的表达。结果：第7、14、21天UUO组中NF-κB表达较假手术组高(P＜0.05)，治疗组在第7天较UUO组低(P＜0.05)。结论：温肾活血化湿方可能通过抑制NF-κB的过度表达从而延缓肾间质纤维化的进程。
Abstract:
Objective: To reveal the mechanism of WenShen HuoXue HuaShi prescription in preventing and treating renal interstitial fibrosis initially by exploring its influence on the expressions of nuclear factor-κB (NF-κB) in renal tissue of unilateral ureteral obstruction (UUO) induced rat model with renal interstitial fibrosis. Methods: Rat model with renal interstitial fibrosis was established by UUO method, eighty rats were separated into sham operation group, UUO group, losartan group and WenShen HuoXue HuaShi prescription group, 20 rats each group; they began to lavage from the second day after successfully establishing the models, the rats in the sham operation group and UUO group accepted intragastric administration of physiological saline in the same amounts, losartan group losartan [10 mg/(kg·d)], WenShen HuoXue HuaShi prescription group WenShen HuoXue HuaShi decoction [9.8 mg/(kg·d)]. In the seventh, 14th and 21st day after successful establishing the models, renal tissue from the obstructed kidney were taken out and dyed by HE and Masson, to observe pathological changes of renal tissue, the expressions of NF-κB were detected in renal tissue by immunohistochemical method. Results: The expressions of NF-κB in UUO group were higher than these of sham operation group in the seventh, 14th and 21st day (P＜0.05), the treatment group was lower than UUO group in the expressions of NF-κB at the seventh day (P＜0.05). Conclusion: WenShen HuoXue HuaShi prescription could delay the progression of renal interstitial fibrosis by inhibiting over expressions of NF-κB possibly.
