摘要:
目的：探讨丹参酮ⅡA对人宫颈癌Hela细胞增殖与凋亡的影响。方法：用梯度浓度(12.5、25、50、100和200 μg/mL)丹参酮ⅡA处理人宫颈癌Hela细胞，MTT法检测丹参酮ⅡA对细胞增殖的影响，Western blotting法检测JAK2/STAT3信号通路相关蛋白p-JAK2、JAK2、p-STAT3和STAT3表达。RT-PCR法检测JAK2/STAT3信号通路相关凋亡基因bax、bcl-2表达。结果:100和200 μg/mL浓度丹参酮ⅡA对Hela细胞增殖具有极显著抑制作用，与对照组比较，100和200 μg/mL浓度处理的细胞，蛋白p-JAK2、p-STAT3均降低，蛋白JAK2、STAT3变化不显著；基因bcl-2表达降低，bax表达升高。结论：100和200 μg/mL浓度的丹参酮ⅡA通过JAK2/STAT3信号通路影响Hela细胞的增殖与凋亡。
Abstract:
Objective: To elucidate the effects of tanshinoneⅡA on the proliferation and apoptosis of human cervical cancer cells (Hela). Methods: Hela were treated with tanshinone Ⅱ A at the gradient concentrations of 12.5, 25, 50, 100 and 200 μg / mL, MTT method was used to detect its effects on cellular proliferation, Western blotting to measure the expressions of JAK/STAT3 signaling pathway related protein p-JAK2, JAK2, P-STAT3 and STAT3. RT-PCR to measure the expressions of JAK/STAT3 signaling pathway apoptosis related protein bax and bcl-2. Results: Tanshinone ⅡA at the concentrations of 100 and 200 μg / mL could significantly inhibit the proliferation of Hela cells, compared with the control group, p-JAK2 and p-STAT3 protein reduced, the changes of JAK2 and STAT3 protein were insignficant in the cells processed with 100 and 200 μg / mL; the expressions of bcl-2 lowered while bax gene expressions rose. Conclusion: Tanshinone ⅡA at the concentrations of 100 and 200 μg / mL can affect the proliferation and apoptosis of Hela cells via JAK2 / STAT3 signaling pathway.
