摘要:
目的探究壮药青风藤总生物碱对类风湿关节炎模型大鼠血清P-选择素（P-selectin，CD62P）、血小板内皮细胞黏附因子（platelet endothelial cell adhesion molecule 1，PECAM-1）表达的影响。 方法选用40只SD大鼠，分为空白组，模型组，青风藤低、中、高剂量组，每组8只。除空白组外，其余各组均使用Ⅱ型胶原酶诱导关节炎模型。各给药组给予对应药物灌胃，给药4周后收集大鼠血清，采用ELISA法检测血清中P-选择素、PECAM-1的表达。 结果造模后，造模组大鼠关节肿胀明显，血清中P-选择素、PECAM-1表达明显高于空白组（P＜0.05）。给药后，与模型组比较，青风藤各剂量组大鼠血清中P-选择素、PECAM-1表达均明显降低（P＜0.05），且青风藤高剂量组明显低于青风藤低剂量组（P＜0.05）。 结论壮药青风藤总生物碱能降低模型大鼠血清中P-选择素及PECAM-1的表达，从而减轻类风湿关节炎症状。
Abstract:
ObjectiveTo survey the effects of Zhuang medicine total alkaloids of Qingfengteng (sinomenii caulis) on the expressions of serum P-selectin (CD62P) and PECAM-1 in rats with rheumatoid arthritis (RA). MethodsForty SD rats were chosen and randomized into the blank group, the model group, low, moderate and high doses groups of Qingfengteng, eight rats in each group. Except the blank group, the rats in other groups were prepared into arthritis models by using type Ⅱ collagenase. All the medication administration groups were drenched with the corresponding medicine, after medicating for four weeks and collecting the serum of the rats, ELISA method was used to detect the expressions of CD62P and PECAM-1 in the serum. ResultsAfter modeling, joint swelling of the rats in the modeling group was remarkable, and the expressions of P-selectin and PECAM-1 in the serum were more apparent than these of the blank group (P<0.05). After medication, compared with the model group, the expressions of P-selectin and PECAM-1 in different doses groups of Qingfengteng were decreased notably (P<0.05), and high dose group of Qingfengteng was obviously lower than low dose group of Qingfengteng (P<0.05). ConclusionZhuang medicine total alkaloids of Qingfengteng could lower the expressions of P-selectin and PECAM-1 in the serum of rat models, consequently to relieve the symptoms of rheumatoid arthritis.
