摘要:
目的研究理冲汤对性激素诱导的子宫肌瘤大鼠Wnt/β-catenin信号通路的调节作用。 方法将60只大鼠随机分为对照组、模型组、理冲汤组和阳性对照组，每组15只，除对照组外，其余大鼠采用苯甲酸雌二醇联合孕酮建立子宫肌瘤大鼠模型，理冲汤组大鼠按照4.5 mL/kg的剂量灌胃给予大鼠理冲汤，阳性对照组大鼠按照2.25 mg/kg的剂量灌胃给予大鼠米非司酮，对照组及模型组灌胃给予大鼠等量生理盐水，每日1次，连续给药28天。检测大鼠血清雌二醇及孕酮水平、子宫系数、子宫组织病理学变化及病理学评分、子宫平滑肌厚度、子宫wnt7a及β-catenin mRNA和蛋白水平。 结果与模型组比较，理冲汤组大鼠血清雌二醇及孕酮水平、子宫系数、子宫平滑肌厚度及病理学评分、子宫β-catenin mRNA和蛋白水平均显著降低（P＜0.05）、子宫wnt7a mRNA和蛋白水平显著升高（P＜0.05）；大鼠子宫组织病理学明显改善。 结论理冲汤能够显著降低大鼠性激素水平及子宫系数，改善大鼠组织病理学变化，其机制可能与调节Wnt/β-catenin信号通路有关。
Abstract:
ObjectiveTo study the regulatory effects of Lichong decoction on Wnt/β-catenin signal pathway in rats with hysteromyoma induced by sex hormone. MethodsAltogether 60 rats were randomly divided into control group, model group, Lichong decoction group and positive control group, with 15 rats in each group. Except for the control group, the other rats were treated with estradiol benzoate combined with progesterone to establish the model of hysteromyoma. The rats in the Lichong decoction group were given Lichong decoction at a dose of 4.5 mL/kg by gavage, and the rats in the positive control group were given mifepristone at the dose of 2.25mg/kg. The rats in the control group and model group were given the same amount of normal saline, once each day, consecutively medicating for 28 days. To detect the levels of serum estradiol and progesterone, uterine coefficient, histopathologic changes in the uterus and pathological scores, uterine smooth muscle thickness, the levels of uterine wnt7a and β-catenin mRNA and protein in rats. ResultsCompared with the model group, the levels of serum estradiol and progesterone, uterine coefficient, uterine smooth muscle thickness, pathological scores, the levels of uterine β-catenin mRNA and protein decreased noticeably in Lichong decoction (P<0.05), the levels of uterine wnt7a mRNA and protein increased (P<0.05); the uterine histopathology of rats was improved significantly. ConclusionLichong decoction could remarkably lower the levels of sex hormone and uterine coefficient, and improve uterine histopathology of rats, and its mechanism might be related to regulating Wnt/β-catenin signal pathway.
