摘要:
目的探讨解毒方对大鼠肝星状细胞（rat hepatic stellate cells，HSCs）HSC-T6细胞增殖及凋亡的影响。 方法将SPF级SD大鼠随机分为空白组、解毒方高剂量组及解毒方低剂量组，每组5只。解毒方高、低剂量组分别给予解毒方42、8.4 g/（kg·d）剂量灌胃，空白组给予等体积生理盐水，干预3天后腹主动脉取血制备含药血清，用含药血清培养活化的HSC-T6细胞，利用MTT法检测细胞增殖情况、流式细胞术检测HSC-T6细胞凋亡情况，并用蛋白免疫印迹法检测凋亡相关蛋白［半胱天冬氨酸蛋白酶3（cysteinyl aspartate specific proteinase 3，Caspase-3）、Caspase-7、Caspase-9、B淋巴细胞瘤（B-cell lymphoma-2，Bcl-2）、BCL2-AssociatedX的蛋白质（BCL2-AssociatedX，BAX）、磷酸化c-Jun氨基末端激酶（c-Jun N-terminalkinase，P-JNK）］表达情况。 结果低剂量解毒方含药血清对HSC-T6细胞增殖无明显抑制作用，而高剂量解毒方含药血清能够明显抑制HSC-T6细胞增殖（P＜0.05）；低、高剂量解毒方含药血清均能够诱导HSC-T6细胞凋亡，能够诱导Caspase-3、Caspase-7、Caspase-9活化，抑制Bcl-2蛋白表达，上调BAX蛋白表达，诱导JNK磷酸化。 结论解毒方含药血清能够抑制肝星状细胞增殖和凋亡，诱导肝星状细胞凋亡的作用可能与改变Bcl-2/Bax比例、激活Caspase级联反应及JNK磷酸化有关。
Abstract:
ObjectiveTo explore the effects of Jiedu formula on the proliferation and apoptosis of rat HSC-T6 cells. MethodsSPF-grade SD rats were randomly assigned to the blank group, high and low doses groups of Jiedu formula with five in each. High and low doses groups of Jiedu formula were administered Jiedu formula via oral gavage at dose of 42 and 8.4 g/(kg·d), respectively, and the blank group received an equal volume of normal saline, drug-containing serum was prepared from blood collected via the abdominal aorta after three days of intervention, and employed to cultivate activated HSC-T6 cells. MTT method was applied to detect cellular proliferationm, flow cytometry to measure the apoptosis of HSC-T6 cells, and Western blotting to determine the expressions of apoptosis-related proteins including Caspase-3, Caspase-7, Caspase-9, Bcl-2, BAX and P-JNK. ResultsLow-dose JDF-containing serum showed no significant inhibitory effects on the proliferation of HSC-T6 cells, while high-dose JDF-containing serum could obviously suppress the proliferation of HSC-T6 cells (P<0.05); low and high-dose JDF-containing serum could induce the apoptosis of HSC-T6 cells, activate Caspase-3, Caspase-7 and Caspase-9, and inhibit the expressions of Bcl-2 protein, upregulate the levels of BAX protein and induce JNK phosphorylation. ConclusionDrug-containing serum could inhibit the proliferation and apoptosis of rat HSC-T6 cells, and the effects of inducing apoptosis in HSC-T6 cells may be related to the alteration of Bcl-2/Bax ratio, the activation of the Caspase cascade and JNK phosphorylation.
