摘要:
目的:探讨黄芪、小蓟通过NF-κB信号通路对大鼠肾小球系膜细胞的影响。方法:体外培养大鼠肾小球系膜细胞,用血管紧张素Ⅱ刺激系膜细胞增生,分别给予黄芪、小蓟,用酶联免疫吸附(ELISA)法检测肾小球系膜细胞上清液NF-κB和FN的表达。结果:与正常组比较,模型组和黄芪高、低剂量组,小蓟高、低剂量组大鼠肾小球系膜细胞中NF-κB和FN表达均升高且差异有统计学意义(P<0.05);与模型组比较,黄芪高、低剂量组、小蓟高、低剂量组均可降低NF-κB信号转导通路中NF-κB及FN的表达,差异有统计学意义(P<0.05)。结论:黄芪、小蓟可能通过下调肾小球系膜细胞NF-κB和FN蛋白表达的水平,从而调整核因子信号转导通路的兴奋性,减轻肾小球的损伤,减缓肾小球的硬化。
Abstract:
Objective: To discuss the influence of HuangQi (Astragalus mongholicus) and XiaoJi (Cirsium setosum) on mesangial cells of the rats by NF-κB signal channel. Methods: Mesangial cells of the rats were cultivated in vitro, the proliferation of mesangial cells were stimulated by angiotensinⅡ, the expressions of NFκB and FN in liquid supernatant of mesangial cells were detected by ELISA method after giving HuangQi and XiaoJi respectively. Results: Compared with the normal group, the expressions of NF-κB and FN in liquid supernatant of mesangial cells raised in the model group, high and low dosage groups of HuangQi, high and low dosage groups of XiaoJi, and there were significant differences (P<0.05); compared with the model group, high and low dosage of HuangQi, high and low dosage of XiaoJi could decrease the expressions of NF-κB and FN in NF-κB signal channel, and the difference showed statistical meaning (P<0.05). Conclusion: HuangQi and XiaoJi could lower the excitability of nuclear factor signal channel, lighten glomerular injury and slow down glomerular sclerosis by downregulating the expressions of NF-κB and FN of mesangial cells.
