摘要:
目的:优选金荞麦有效成分的提取纯化工艺。方法:以表儿茶素(Epicatechin)的提取率、吸附率和解吸附率为指标,结合HPLC含量测定,选择最佳提取条件、树脂型号和纯化工艺;同时考察乙酸乙酯和正丁醇的纯化效果。结果:最佳超声提取工艺条件为乙醇浓度50%,液料比1∶10,提取3次,30 min/次;最佳纯化工艺为上样液浓度0.5 g/mL,pH5~6,以3 BV/h流过D101大孔树脂,流出液重复上柱2次,加4 BV水洗脱除杂,加3 BV 50%乙醇洗脱富集有效成分;提取液中有效成分也可用正丁醇萃取富集。结论:采用乙醇超声提取,D101大孔树脂分离纯化或正丁醇纯化金荞麦有效成分的工艺方法稳定、可行。
Abstract:
Objective: To optimize the extraction and purification technology for the active ingredients of JinQiaoMai[Fagopyrum dibotrys (D. Don) Hara]. Methods: The purification technology, the types of resins and the best extraction conditions were chosen by taking the extraction rate, absorption rate and desorption rate of epicatechin as the indexes combined with content determination by HPLC. Results: The best ultrasonic extraction conditions were the concentrations of ethanol 50%, liquid-material ratio was one to ten, extraction times were three times, 30 min each time; the best purification technology:the concentrations of sample solution were 0.5 g/mL, pH of 5 to 6, absorbed by macroporous resin with a flow rate of 3 BV/h, effluent liquid on column twice repeatedly, adding 4BV of water to elute and remove impurities, adding 3 BV of 50% ethanol to elute the enriched active ingredients; the active ingredients in the extract could be leached and enriched by n-butyl alcohol. Conclusion: Active ingredients of JinQiaoMai are extracted by ethanol ultrasonic extraction, separated and purified by D101 macroporous resin or purified by n-butyl alcohol, and the technology method is stable and feasible.
