摘要:
目的:研究甘草酸抑制中波紫外线(UVB)诱导人皮肤角质形成细胞(HaCaT)凋亡的机制。方法:以不同浓度的甘草酸作用于人皮肤角质形成细胞(HaCaT细胞),噻唑蓝比色(MTT)法筛选药物的有效安全浓度;用30 mJ/cm2的UVB照射HaCaT细胞,制备光老化模型,MTT 法检测细胞增殖率;流式细胞仪检测各组细胞总凋亡率;实时定量PCR(RT-PCR)法检测Bax、Bcl-2 mRNA表达水平变化;Western Blot 法检测各组细胞上清中 Bax、Bcl-2蛋白表达量。结果:1×10-7、1×10-6、1×10-5 mol/L甘草酸为最佳有效安全浓度。与空白组比较,UVB组细胞增殖率显著降低(P<0.01);细胞总凋亡率显著显著升高(P<0.01);Bax mRNA表达水平显著升高,Bcl-2 mRNA表达水平显著降低(P<0.01); Bax蛋白表达量升高,Bcl-2蛋白表达量显著降低(P<0.01)。与UVB组比较, 1×10-7、1×10-6、1×10-5 mol/L甘草酸+UVB组细胞增殖率显著升高(P<0.01);细胞总凋亡率显著降低(P<0.01);Bax mRNA表达水平显著降低,Bcl-2 mRNA表达水平显著升高(P<0.01,P<0.05); Bax蛋白表达量显著降低,Bcl-2蛋白表达量显著升高(P<0.01,P<0.05)。结论:甘草酸能抑制UVB诱导的HaCaT细胞凋亡,其机制与其下调促凋亡基因Bax mRNA和蛋白的表达,上调抑凋亡基因Bcl-2 mRNA和蛋白的表达有关。
Abstract:
Objective:To research on the mechanism of glycyrrhizic acid inhibiting the apotosis of HaCaT induced by ultraviolet B(UVB). Methods: Different concentrations of glycyrrhizic acid were acted on HaCaT cells, MTT method was used to screen the effective safe concentrations of the drugs; HaCaT cells were irradiated with 30 mJ/cm-2 UVB to set up light aging models, MTT method was used to detect cellular proliferation rate; flow cytometry was adopted to detect total apoptosis rate of the cells in different groups; real-time quantitative PCR (RT-PCR) method to detect the expressions of Bax and Bcl-2 mRNA; Western Blot method to measure the expression of Bax and Bcl-2 protein in supernatant fluid of the cells in different groups. Results: 1×10-7, 1×10-6 and 1×10-5 mol/L glycyrrhizic acid was the best effective safe concentrations. Compared with the blank group, cellular proliferation rates of UVB group decreased notably(P<0.01); total apoptosis rate of the cells increased significantly (P<0.01); the levels of Bax mRNA expressions rose notably, the expressions of Bcl-2 mRNA lowered clearly (P<0.01); the expressions of Bax protein increased notably, the expression of Bcl-2 decreased obviously (P<0.01). Compared with UVB group, cellular proliferation rate increased clearly in 1×10-7, 1×10-6 and 1×10-5 mol/L glycyrrhizic acid+UVB group (P<0.01); total apoptosis rate of the cells lowered notably (P<0.01); the expressions of Bax mRNA lowered apparently, the expression of Bcl-2 mRNA raised notably (P<0.01, P<0.05); the expressions of Bax protein decreased, the expressions of Bcl-2 protein increased obviously (P<0.01, P<0.05). Conclusion:Glycyrrhizic acid could inhibit the apoptosis of HaCaT cells induced by UVB, its mechanism might be related to down regulating the expressions of Bax mRNA and protein, up regulating the expressions of Bcl-2 mRNA and protein.
