摘要:
目的:建立前胡配方颗粒含量测定及HPLC指纹图谱研究方法,为前胡配方颗粒的质量评价提供依据。方法: 采用HPLC方法测定白花前胡甲素(praeruptorin A)和白花前胡乙素(praeruptorin B)的含量,采用Sepax HP C18色谱柱(4.6 mm×250 mm,5 μm),流动相:甲醇-水(75∶25),检测波长:321 nm,柱温:25℃,建立HPLC指纹图谱。结果:白花前胡甲素进样量在11.230~179.680 mg/L峰面积呈良好线性关系(R2=1.000 0),标准曲线为:Y=22.4231X+4.2344,平均加样回收率为97.58%(RSD=2.81%, n=9);白花前胡乙素进样量在5.652~90.434 mg/L峰面积呈良好线性关系(R2=1.000 0),标准曲线为:Y=20.737 0 X+0.936 6,平均加样回收率为101.15%(RSD=1.82%,n=9)。建立了前胡配方颗粒指纹图谱,通过HPLC指纹图谱分析确定前胡配方颗粒共有模式图,10批前胡配方颗粒样品指纹图谱与对照指纹图谱相似度均>0. 97,相似度高,可实现前胡配方颗粒化学成分整体评价。结论:本研究所采用定性定量方法稳定可行,可用于前胡配方颗粒的质量控制。
Abstract:
Objective: To provide the reference for quality evaluation of Qianhu dispensing granules by establishing the research methods of determining the contents of Qianhu dispensing granules and HPLC fingerprint method. Methods: HPLC method was used to determine the contents of praeruptorin A and praeruptorin B, Sepax HP C18 chromatographic column (4.6 mm×250 mm, 5 μm), mobile phase: methyl-water (75:25), detection wavelength:321nm, column temperature:25℃, to establish HPLC fingerprint. Results: Praeruptorin A showed better linear relationship when the sample size was in the range between 11.230 and 179.680 mg/L (R2=1.000 0), standard curve was: Y=22.423 1 X+4.234 4, average recovery rate was 97.58% (RSD=2.81%, n=9); praeruptorin B presented better linear relationship when the sample size was in the range between 5.652 and 90.434 mg/L(R2=1.000 0), standard curve was: Y=20.737 0 X+0.936 6, average recovery rate was 101.15% (RSD=1.82%, n=9); the fingerprint of Qianhu dispensing granules was established, the common model diagram of Qianhu dispensing granules was ensured by analyzing HPLC fingerprint, the similarities of ten batches of samples and the control group were>0.97, high similarity, the whole assessment of chemical ingredients of Qianhu dispensing granules could be realized. Conclusion: The qualitative and quantitative method is stable and feasible, and it could be used for quality control of Qianhu dispensing granules.
