摘要:
目的研究蒙药蓝刺头对绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)的防治作用,阐述其在蒙医“补暖补精,固骨质壮骨”理论下促进骨形成、抑制破骨的机制。 方法取Wistar大鼠77只,随机分为蓝刺头高、中、低剂量组,强骨胶囊组,己烯雌酚组,模型组,假手术组,每组11只。通过去卵巢复制PMOP模型,用蒙药蓝刺头、强骨胶囊和己烯雌酚分别灌胃模型大鼠90天,观察比较各组大鼠血清转化生长因子β1(transforming growth factor-β1,TGF-β1)、骨密度(bone mineral density,BMD)、子宫病理学变化情况。 结果与模型组比较,中剂量的蒙药蓝刺头能有效防治绝经后骨质疏松症模型大鼠骨质疏松症,同时明显提高绝经后骨质疏松症模型大鼠血清TGF-β1水平(P<0.05)。子宫HE染色切片分析显示:中剂量的蒙药蓝刺头能显著改善大鼠去卵巢后对于子宫的影响,子宫肌全层明显增厚、内膜腺体内部的直径明显增大、内膜腺体腔内面积所占视野百分比明显增大、内膜上皮深度明显增大(P<0.05)。 结论蒙药蓝刺头具有提高去势大鼠BMD作用,一定程度上抑制了雌激素缺乏导致的子宫功能的丧失,其通过雌激素经典通路对于PMOP有防治作用。
Abstract:
ObjectiveTo illustrate the mechanism of promoting bone regeneration and inhibiting bone destroying guided by the calssical theory of mongolian medicine by studying the effects of mongolian medicine Lancitou on postmenopausal osteoporosis (PMOP) via estrogen classical signaling pathway. MethodsAll 77 Wistar rats were chosen and randomized into high, moderate and low doses groups of Lancitou, Qianggu capsules group, diethylstilbestrol (DES) group, the model group and sham operation group, 11 rats each group. PMOP models were established by ovariectomizing; rat models were drenched with mongolian medicine Lancitou, Qianggu capsules and DES for 90 days, to observe and compare serum transforming growth factor β1(TGF- β1), bone mineral density (BMD) and uterine pathology of the rats in different groups. ResultsCompared with the model group, moderate dose of mongolian medicine Lancitou could effectively prevent osteoporosis of PMOP rats from BMD, and simultaneously lift TGF-β1 levels obviously (P<0.05). The analysis of uterine slice after HE staining showed: moderate dose of mongolian medicine Lancitou could remarkably improve ovariectomizing influence on the uterus, total uterine muscle thickened notably, internal diameter of endometrial glands enlarged, visual field percentage of endometrial glands cavity area and endometrial epithelial depth increased notably (P<0.05). ConclusionMongolian medicine Lancitou could raise BMD of ovariectomized rats, inhibit the loss of uterine function caused by estrogen to a certain extent, and it could effectively prevent and treat PMOP through estrogen classical signaling pathway.
