摘要:
目的探讨木丹颗粒对糖尿病周围神经病变(diabetic peripheral neuropathy,DPN)大鼠坐骨神经Toll样受体4(toll-like receptor-4,TLR4)蛋白相关通路的调控作用。 方法将SD雄性大鼠随机分为空白对照组和高糖组,高糖组制备糖尿病模型,并分为模型组,α-硫辛酸组和木丹颗粒组,给予相应药物干预12周后,收集大鼠腹主动脉血制备血清,采用酶联免疫吸附剂法(enzyme linked immunosorbent assay,ELISA)测定肿瘤坏死因子α(tumor necrosis factor α,TNF-α)和白细胞介素6(interleukin 6,IL-6)含量,收集坐骨神经用于RT-PCR法、免疫荧光化学法和Western blot法测定Toll样受体4(toll-like receptor 4,TLR4)、髓样分化因子88(myeloid differentiation factor 88,MyD88)、核因子κB(nuclear factor-κB,NF-κB)和p38丝裂原活化蛋白激酶(p38 mitogen activated protein kinase,p38 MAPK)的mRNA和蛋白表达,电镜下观察大鼠主骨神经髓鞘结构形态。 结果与空白对照组比较,模型组大鼠神经传导速度(nerve conduction velocity,NCV)显著降低(P<0.05),鼠尾热痛阈值(thermal pain threshold,TPT)显著升高(P<0.05),坐骨神经出现脱髓鞘现象;血清中炎症因子TNF-α和IL-6含量显著升高(P<0.05),坐骨神经中TLR4、MyD88、NF-κB和p38 MAPK的mRNA和蛋白表达显著升高(P<0.05);与模型组比较,木丹颗粒组大鼠NCV显著升高(P<0.05),TPT显著降低(P<0.05);脱髓鞘现象显著改善;血清中炎症因子TNF-α和IL-6含量显著降低(P<0.05),坐骨神经中TLR4、MyD88、NF-κB和p38 MAPK的mRNA和蛋白表达均显著降低(P<0.05)。 结论木丹颗粒能够通过调控TLR4/MyD88/NF-κB通路和TLR4/p38 MAPK通路改善炎症反应,改善周围神经功能的损伤,从而防治DPN。
Abstract:
ObjectiveTo discuss the regulatory effects of Mudan granules on TLR4 protein related pathway in sciatic nerve of DPN rats. MethodsSD male rats were randomized into blank control group and high glucose group, high glucose group were prepared into DM models, and divided into the model group,α-lipoic acid group and Mudan granules group, after 12 weeks of drug intervention, abdominal aortic blood was collected to prepare the serum, ELISA method was used to detect the contents of TNF-α and IL-6, after collecting sciatic nerve, RT-PCR, immunofluorescence method and Western blot method were applied to measure the expressions of TLR4, MyD88, NF-κB and p38 MAPK mRNA and protein, the myelin sheath structure of rat main bone nerve was observed under electron microscrope. ResultsCompared with blank control group, NCV of the rats in the model group lowered noably (P<0.05), TPT of rat tail rose remarkably (P<0.05), demyelination of sciatic nerve occurred; the contents of TNF-α and IL-6 in the serum increased notably (P<0.05), the expressions of TLR4, MyD88, NF-κB and p38 MAPK mRNA and protein rose apparently (P<0.05); compared with the model group, NCV of the rats in Mudan granules group lifted notably (P<0.05), TPT reduced signficantly (P<0.05); demyelina-tion improved notably, the contents of TNF-α and IL-6 in the serum lowered notably (P<0.05), the expressions of TLR4, MyD88, NF-κB and p38 MAPK mRNA and protein in sciatic nerve decreased notably (P<0.05). ConclusionMudan granules could improve inflammatory reaction, the injury of peripheral nerve function through regulating TLR4/MyD88/NF-κB pathway and TLR4/p38 MAPK pathway, therefore to prevent and treat DPN.
