摘要:
目的利用网络药理学方法探讨金铃子散治疗原发性痛经的作用机制。 方法通过TCMSP检索金铃子散中两味中药的化合物,筛选出有效成分及对应靶点;利用人类基因(GeneCards)、在线人类孟德尔遗传(online mendelian inheritance in man,OMIM)数据库、药物靶点(therapeutic target database,TTD)数据库获取疾病靶点并构建蛋白质-蛋白质相互作用关系(protein-protein Interaction,PPI);利用David数据库进行基因功能(gene ontology,GO)和基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)分析;运用Cytoscape软件构建金铃子散“活性成分-靶点-通路”网络。 结果得到金铃子散的有效成分55种,作用靶点207个,疾病靶点683个;得到交集靶点59个,将交集靶点上传到STRING数据库得到由59个节点、155条边构成的平均节点度值为5.25、平均局部聚类系数为0.518的“药物-疾病”PPI;GO分析得到156个生物过程、15个细胞组分、40个分子功能;KEGG分析得到53条信号通路,取前20位绘制气泡图。 结论金铃子散可能通过多成分、多靶点、多通路实现治疗原发性痛经的目的。
Abstract:
ObjectiveTo discuss the effects of Jinlingzi powder in the treatment of primary dysmenorrhea by using network pharmacology. MethodsThe compounds of two herbs in Jinlingzi powder were searched via TCMSP, to screen out the active ingredients and the corresponding targets; GeneCards, OMIM database and TTD databases were utilized to obtain disease targets and construct PPI; David database was applied to perform GO and KEGG analysis; Cytoscape software was used to build "the active ingredients-targets-pathway" network of Jinlingzi powder. ResultsWe have obtained 55 active ingredients from Jinlingzi powder, 207 targets and 683 disease targets; after uploading 59 intersection targets to the STRING database, PPI of "medicine-disease" was composed of 59 nodes and 155 edges with an average node degree value of 5.25 and an average local clustering coefficient of 0.518. GO analysis yielded 156 biological processes, 15 cellular components and 40 molecular functions; KEGG analysis involved 53 signaling pathways, and the first 20 ones were chosen to draw the bubble chart. ConclusionJinlingzi powder could treat primary dysmenorrhea possibly through many ingredients, targets and pathways.
