目的利用网络药理学研究方法探讨薄荷治疗深阿尔茨海默病(Alzheimer′s disease,AD)形成的作用机制。 方法利用 (traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)数据库,挖掘薄荷有效成分并预测其作用靶点,通过GeneCards、OMIM等数据库筛选AD形成的靶标。运用Cytoscape 软件构建薄荷治疗AD形成的“成分-靶标-通路-疾病”网络,通过STRING数据库构建靶点蛋白相关作用网络并进行关键靶点的生物功能注释及通路分析。 结果薄荷10个有效成分通过干预82个靶点、144条通路发挥治疗AD形成的作用。 结论薄荷有效成分可能通过抑制乙酰胆碱、细胞免疫、血管内皮因子反应等药理作用治疗AD形成。
Abstract:
ObjectiveTo discuss the mechanism of action of Bohe (Menthae haplocalycis herba) in treating Alzheimer's disease (AD) by using the research methods of network pharmacology. MethodsTCMSP was utilized to excavate the active ingredients of Bohe and predict its targets, AD targets were screened by GeneCards and OMIM databases. Cytoscape software was applied to construct the network of "ingredient-target-pathway-disease" for treatment of AD by Bohe, STRING database was used to build the network of target-protein interaction, perform biological function annotation of key targets and analyze the pathways. ResultsBohe contains ten effective constituents, and it exerts theraputic effects on AD by intervening 82 targets and 144 pathways. ConclusionActive ingredients of Bohe treat AD possibly through inhibiting acetyl choline (ACH), cellular immunity and vascular endothelial factor response.
