摘要:
目的考察乌梅总黄酮(Fructus Mume total flavone,FMF)对6-OHDA诱导帕金森病(Parkinson?s disease,PD)大鼠脑线粒体自噬的影响。 方法选择100只雄性SD大鼠,设假手术组15只,另外85只大鼠采用6-OHDA(5 μg/μL)左侧纹状体注射制备PD大鼠模型。造模成功后将大鼠随机分成模型组,FMF低、中、高剂量组,每组15只。FMF低、中、高剂量组分别给予40、80、160 mg/kg FMF灌胃给药,假手术组及模型组给予相等容积生理盐水。干预4周,利用电镜观察大鼠脑组织线粒体形态、结构,采用比色法检测线粒体ATP酶活性,采用WB实验测定线粒体自噬相关蛋白LC3-Ⅱ、Beclin-1、BNIP3的表达。 结果与假手术组相比,模型组大鼠脑组织线粒体明显肿胀,线粒体嵴断裂,部分溶解、消失,排列紊乱;Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶活力明显下降(P<0.05);LC3-Ⅱ、Beclin-1、BNIP3蛋白含量明显下降(P<0.05)。与模型组相比,FMF各剂量治疗组大鼠脑组织线粒体结构破坏呈剂量依赖性减轻,线粒体结构尚完整,部分线粒体稍肿胀,线粒体嵴排列较整齐,线粒体嵴的缺失较少,尤以高剂量组线粒体保护作用最明显。FMF各剂量组Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶活性明显提高(P<0.05),LC3-Ⅱ、Beclin-1、BNIP3蛋白含量明显升高(P<0.05)。 结论FMF能够显著修复6-OHDA诱导PD大鼠的脑线粒体功能损伤,FMF参与调控6-OHDA诱导的脑组织损伤中线粒体自噬水平,促进线粒体自噬。
Abstract:
ObjectiveTo investigate the influence of Fructus Mume total flavone (FMF) on mitophagy in 6-OHDA-induced PD rat. MethodsAll 100 male SD rats were chosen, 15 ones were set as sham operation group, the rest 85 rats were prepared into PD models by left striatum injection of 6-OHDA, they were randomly allocated into the model group, low, moderate and high doses groups of FMF after successfully modeling, 15 rats in each group. Low, moderate and high doses groups of FMF accepted intragastric administration of FMF, 40, 80 and 160mk/kg. Sham operation group and the model group were given the equivalent volume of normal saline. After intervention for four weeks, to observe the morphology and structure of mitochondria in cerebral tissue by using electron microscope. To detect the activity of mitochondrial ATP enzyme by using colorimetry, and to measure the expressions of mitochondrial autophagy related proteins LC3-Ⅱ, Beclin-1 and BNIP3 by WB experiments. ResultsCompared with sham operation group, the mitochondria in cerebral tissue are obviously swollen, mitoch-ondrial cristae is broken, partially dissolved, disappeared and disordered; the activity of Na+-K+-ATP enzyme and Ca2+-Mg2+-ATP was remarkably reduced (P<0.05); the contents of LC3-Ⅱ, Beclin-1 and BNIP3 were lowered apparently (P<0.05). Compared with the model group, the damage of mitochondrial structure in cerebral tissue was relieved in dose-dependent manner in different doses groups of FMF, the mitochondrial structure is still intact, some mitochondria are slightly swollen, the mitochondrial cristae are arranged neatly, the protective effects were the most obvious, especially in high dose group. The activity of Na+-K+-ATP enzyme and Ca2+-Mg2+-ATP was raised remarkably in different doses groups of FMF (P<0.05), the contents of LC3-Ⅱ, Beclin-1 and BNIP3 were lifted markedly (P<0.05). ConclusionFMF could significantly repair brain mitochondrial function damage in PD rats induced by 6-OHDA, participating in the regulation of mitochondrial autophagy induced by 6-OHDA in cerebral tissue damage and promoting mitochondrial autophagy.
