目的运用网络药理学方法探讨稳心颗粒治疗心律失常的作用靶点及通路。 方法对收集到的稳心颗粒相关蛋白靶点和心律失常相关基因靶点进行hub基因筛选、基因本体论功能富集分析(gene ontology,GO)和KEGG富集分析(kyoto encyclopedia of genes and genomes,KEGG)。 结果排名前10的基因分别是血管内皮生长因子A(vascular endothelial growth factor A,VEGFA)、丝氨酸/苏氨酸特异性蛋白激酶1(serine/threonine kinase 1,Akt1)、TP53基因(tumor protein P53,TP53)、Jun proto-oncogene(JUN)、白细胞介素6(interleukin-6,IL-6)等。GO富集显示199个生物过程、44个分子功能、29个细胞成分,涉及保护离子通道、抗氧化及抗炎等方面。KEGG通路显示95条通路,其中31条通路与冠心病相关,分别是PI3K-Akt信号通路、HIF-1信号通路、FoxO信号通路、Thyroid hormone信号通路等。 结论稳心颗粒治疗心律失常具有多成分、多靶点的特点,可能通过保护离子通道、抗氧化及保护心肌细胞等发挥作用。
Abstract:
ObjectiveTo explore the targets and pathways of Wenxin granules in the treatment of arrhythmia by using network pharmacology method. MethodsThe relevant protein targets and arrhythmia related gene targets were collected to conduct hub gene screening, GO and KEGG. ResultsThe top ten genes were VEGFA, Akt1, TP53 gene, JUN, IL-6 and others respectively. GO enrichment analysis showed 199 biological processes, 44 molecular functions, 29 molecular ingredients, involving protecting ion channels, antioxidant and anti-inflammatory aspects. KEGG pathways indicated that there were 95 pathways, among them, 31 pathways were related to CHD, and they were PI3k/Akt signaling pathway, HIF-1 signaling pathway, FoxO signaling pathway, and thyroid hormone signaling pathway respectively. ConclusionWenxin granules have the characteristics of multi-component and multi-target in the treatment of arrhythmia, and it could develop the effects possibly through protecting ion channels, antioxidant and protecting myocardial cells.
