摘要:
目的:研究大蒜素对人舌鳞状细胞癌Tca8113细胞生长的抑制作用及其机制。方法:取对数生长期Tca8113细胞设空白对照组,顺铂组,大蒜素低、中、高剂量组,给药干预48小时后,观察各组细胞生长状态,MTT法检测细胞生长抑制率,流式细胞术检测细胞周期时相变化和细胞凋亡状况,Western blotting法检测Bax、B细胞淋巴瘤-2基因(Bcl-2)、激活型Caspase-3蛋白表达。结果:与空白对照组比较,大蒜素各组和顺铂组Tca8113细胞呈现胞体破裂,核固缩、深染等病理性改变,以大蒜素50 μg/mL组最为显著;大蒜素各组和顺铂组Tca8113细胞增殖抑制率和凋亡率显著提高(P<0.05);大蒜素(25、50 μg/mL)组和顺铂组G2/M期比例显著提高(P<0.05),Bax、激活型Caspase-3蛋白表达显著上调且Bcl-2蛋白显著下调(P<0.05),Bax/Bcl-2值显著提高(P<0.05)。结论:大蒜素对人舌鳞状细胞癌Tca8113细胞生长具有一定的抑制作用,可能与大蒜素阻滞细胞有丝分裂以及调控凋亡相关蛋白表达而促进细胞凋亡有关。
Abstract:
Objective:To study the inhibitory effects of allimin on Tca8113 cellular growth of human tongue squamous cell carcinoma and its mechanism. Methods: Tca8113 cells at logarithmic growth phase were selected and set as blank control group, low, moderate and high doses groups of allimin, and cis-platinum complexes group, after medicating for 48 hours, cellular growth of different groups were observed, the inhibitory rate of cellular growth was detected using MTT method, cellular apoptosis and the phases changes of cellular cycle were measured by flow cytometry, the expressions of Bax, Bcl-2 and activation type Caspse-3 protein were tested using Western blotting method. Results: Compared with blank control group, pathological changes of different groups of allimin and cis-platinum complexes group presented including cell body rupture, karyopyknosis, nuclear hyperchromatism and others, especially the allimin group of 50 μg/mL the most obvious; the apoptotic rate and inhibition ratio of Tca8113 cells in the groups of allimin and cis-platinum complexes group increased obviously (P<0.05); G2/M ratio in allimin groups of 25 and 50 μg/mL and cis-platinum complexes group lifted obviously (P<0.05), the expressions of Bax and activation type Caspse-3 protein rose notably while the expressions of Bcl-2 protein downregulated (P<0.05), Bax/Bcl-2 ratio rose signficantly(P<0.05). Conclusion: Allimin owns certain inhibitory effects to Tca8113 cellular growth of human tongue squamous cell carcinoma, which might be associated to that allimin could promote cellular apoptosis through blocking mitosis and regulating the expressions of apoptosis-related protein.
