目的观察中药复方联合三苯氧胺(tamoxifen,TAM)对MCF-7乳腺癌细胞增殖的MAPK信号转导通路及人类表皮生长因子受体2(human epidermal growth factor receptor,HER-2)基因mRNA表达的影响,探讨中药复方抑制MCF-7乳腺癌细胞增殖的信号传导途径。 方法将造模成功的40只裸鼠随机分为模型组、TAM组、乳结合组(乳岩宁方+TAM)和益结合组(益气养阴方+TAM)。观察各组大鼠一般状态,Western blot法检测乳腺癌组织中ERK1/2、ERα蛋白的表达,RT-PCR法检测乳腺癌组织中HER-2mRNA的表达。 结果1)除模型组外,各用药组荷瘤裸鼠体质量下降,形体消瘦,活动量减少,进食减少,喜蜷卧,以TAM组表现最明显,乳结合组和益结合组状态保持相对较好。2)TAM组、乳结合组和益结合组的ERK1/2、ERα蛋白,HER-2mRNA的表达低于模型组(P<0.01),其中乳结合组的作用最显著,益结合组次之;乳结合组和益结合组与TAM组比较差异有统计学意义(P<0.01);乳结合组和益结合组疗效相当(P>0.05)。 结论中药复方联合TAM可能是通过下调癌组织中HER-2mRNA的表达来抑制MAPK信号通路的活性,从而抑制乳腺癌细胞的增殖,同时中药复方对TAM有协同作用。
Abstract:
ObjectiveTo observe the effects of herbal compound joined with TAM on the expressions of HER-2 mRNA and MAPK signaling transduction pathway in the proliferation of MCF-7 breast cancer cells, and discuss the effects of herbal compound inhibiting signaling transduction pathway of MCF-7 breast cancer cells proliferation. MethodsForty nude mice after modeling successfully were randomized into the model group, TAM group, (Ruyanning prescription+TAM) group and (Qi-benefiting Yin-nourishing prescription+TAM) group. To observe general state of the rats in different groups, to measure the expressions of ERK1/2 and ERɑ protein in the tissue of breast cancer using Western blot method, to measure the expressions of HER-2 mRNA in the tissue of breast cancer using RT-PCR method. Results1)Except the model group, the body mass of tumor bearing nude mice decreased, physical emaciation, the amount of activity decreased, food intake reduced, and they liked to curling up in different administration groups, especially the manifestations of TAM group, the state of the mice in (Ruyanning prescription+TAM) group and (Qi-benefiting Yin-nourishing prescription+TAM) group remained relatively good. 2) The expressions of ERK1/2, ERα protein, HER-2 mRNA of TAM group, (Ruyanning prescription+TAM) group and (Qi-benefiting Yin-nourishing prescription+TAM) group were lower than these of the model group (P<0.01), among them, the effects of (Ruyanning prescription+TAM) group were the most notable, Qi-benefiting Yin-nourishing prescription+TAM group the next; the difference showed statistical meaning when (Ruyanning prescription+TAM) group and (Qi-benefiting Yin-nourishing prescription+TAM) group were compared with TAM group (P<0.01); clinical effects of (Ruyanning prescription+TAM) group and (Qi-benefiting Yin-nourishing prescription+TAM) group were equivalent (P>0.05). ConclusionHerbal compound and TAM may inhibit the activity of MAPK signaling pathway through downregulating the expressions of HER-2 mRNA in tumor tissues, thereby inhibiting the proliferation of breast cancer cells, simultaneously, herbal compound has synergistic effect on TAM.
