摘要:
目的探讨子宫内膜异位症发病机制的关键基因及相应的有效中药。 方法从基因芯片数据库(gene expression omnibus,GEO)下载有关子宫内膜异位症表达的GSE25628(数据集),利用在线GE02R筛选具有显著差异的差异表达基因(differentialy expressed genes,DEGs),并对DEGs进行基因本体(gene ontology,GO)富集分析、京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)通路分析并对差异基因进行蛋白互作;预测关键基因相关的微RNA(microRNAs,miRNAs)并从毒物遗传学数据库(comparative toxicogenomics database,CTD)筛选作用于关键基因的有效中药。 结果从GSE25628数据集中筛选到49个DEGs,其中有5个上调DEGs,44个下调DEGs;DEGs主要参与端粒维护、超分子纤维组织的调节、ERBB2信号通路、蛋白质聚合的调控等生物学过程,存在于细胞核、锥体、细胞间黏附连接处等,与细胞黏附分子、激酶活性等结合;参与调控癌症通路、PI3K-Akt通路、细胞凋亡等通路。预测的关键基因为ATRX、SMC5、TPR、HSP90AA1、HIRA、RAD50、CEP290、PIK3R1;hsa-miR-147、hsa-miR-591、hsa-miR-548s、hsa-miR-410等,可作用于关键基因调控子宫内膜异位症的发展;而木黄酮、白藜芦醇、槲皮素等中药成分可上调关键基因的表达。 结论通过生物信息学筛选出的关键基因及通路,可为进一步研究EMs的发病机制提供思路,也可能是中医药有效成分治疗EMs的关键靶点。
Abstract:
ObjectiveTo explore the key genes of the mechanism of endometriosis and the corresponding effective traditional Chinese medicine. MethodsGSE25628 (data set) related to the expressions of endometriosis was downloaded from GEO, online GEO2R was utilized to screen differentially expressed genes (DEGs) with significant differences, to carry out GO and KEGG of DEGs, and perform protein interaction for the differential genes; micro RNAs related to key genes were predicted, and the effective traditional Chinese medicine acting on key genes were screened. ResultsAll 49 DEGs were screened from GSE25628 data set, among which, there were five up-regulated DEGS and 44 down-regulated DEGs; DEGs mainly participated in the biological processes including telomere maintenance, the regulation of the metamolecular fibrous organization, ERBB2 signaling pathway, the regulation of protein polymerization, they were present in the nucleus, cone, intercellular adhesion junctions, etc, binding with cell adhesion molecules, kinase activity, etc; taking part in cancer pathway, PI3K-Akt pathway and cellular apoptosis. The key genes predicted were ATRX, SMC5, TPR, HSP90AA1, HIRA, RAD50, CEP290, PIK3R1; hsa-miR-147, hsa-miR-591, hsa-miR-548s, hsa-miR-410, acting on the development of endometriosis regulated by key genes; while herbal ingredients such as genitein, resveratrol and quercetin could up regulate the expressions of key genes. ConclusionKey genes and pathways screened out through informatics could provide the thinking for further research on the pathogenesis of EMs, which might be the key targets for the treatment of EMs with active ingredients of Chinese medicine.
