摘要:
目的：建立高效液相色谱法对甘露消毒丹中3种成分（绿原酸、黄芩苷、连翘苷）的分析方法。方法：采用Agilent TC-C18（4.6 mm×250 mm，5 μm）色谱柱，以乙腈（A）-0.1%磷酸水（B）为流动相，梯度洗脱，流速为1.0 mL/min，检测波长为327 nm，0～20分钟后测定绿原酸含量；20分钟以后，230 nm测定黄芩苷和连翘苷含量。结果：甘露消毒丹中3种成分分别为绿原酸在0.0791～1.9775 μg（r=0.999 7），黄芩苷在0.3827～9.5675 μg（r=0.999 9），连翘苷在0.0652～1.630 0 μg（r=0.999 9）范围内线性关系良好；平均回收率分别为98.66%、97.49%和98.52%；平均含量分别为2.389、28.362、4.354 mg/g。结论：该方法准确可靠，分离效果理想，重复性好，可用于甘露消毒丹质量控制。
Abstract:
Objective: To establish the determination method for three ingredients (chlorogenic acid, baicalin and phillyrin) in GanLu XiaoDu pills by HPLC. Methods: Agilent TC-C18 (4.6 mm×250 mm, 5 μm) chromatographic column was used, acetonitrile (A)-0.1% phosphoric acid (B) were taken as the mobile phase, gradient elution, flow rate was 1.0 mL/min, detection wavelength was 327nm, the contents of chlorogenic acid were measured in 0 and 20 minutes, the contents of baicalin and phillyrin were detected in 20 minutes when the detection wavelength was 230nm. Results: The three ingredients of GanLu XiaoDu pills were in the good linear relationship when chlorogenic acid, baicalin and phillyrin were in the ranges of 0.0791～1.9775 μg(r=0.999 7), 0.3827～9.5675 μg(r=0.999 9) and 0.065 2～1.630 0 μg(r=0.999 9); average recovery rate were 98.66%, 97.49% and 98.52% respectively, average contents were 2.389 mg/g, 28.362 mg/g and 4.354 mg/g respectively. Conclusion: The method, accurate, reliable and reproducible, could be used for quality control of GanLu XiaoDu pills.
