摘要:
目的:探讨白藜芦醇对类风湿性关节炎(rheumatoid arthritis,RA)大鼠炎症损伤的保护作用及其对Tenascin-c/TLR4信号通路的影响。方法:选择48只雄性SD大鼠,分为对照组,模型组,雷公藤多苷(20 mg/kg)组,白藜芦醇低、中、高剂量(15、30、60 mg/kg)组,每组8只,连续灌胃给药42天。除对照组外,剩余组大鼠在实验第1、14天2次在大鼠背部、尾根部多点皮内注射牛Ⅱ型胶原与完全弗氏佐剂,制备胶原诱导性关节炎(collagen induced arthritis,CIA)大鼠模型。记录治疗期间各组大鼠体质量、关节肿胀及多发关节炎指数(arthritis indexes,AI)的变化,分析血清炎性因子白细胞介素6(interleukin-6,IL-6)、白介素1β(interleukin-1β,IL-1β)及肿瘤坏死因子α(tumor necrosis factor α,TNF-α)水平,检测滑膜组织Tenascin-c及TLR4蛋白表达情况。结果:与对照组比较,模型组大鼠体质量增长迟缓,后肢关节严重肿胀,AI值显著增高,血清IL-6、IL-1β 及TNF-α水平明显升高,且Tenascin-c及TLR4蛋白表达量也显著升高(P<0.05);与模型组大鼠比较,各药物干预组能够改善模型大鼠体质量、降低关节肿胀率及AI值(P<0.05),可显著抑制炎症因子IL-6、IL-1β 及TNF-α的产生(P<0.05),还能明显抑制炎性通路蛋白Tenascin-c和TLR4表达(P<0.05)。结论:白藜芦醇可通过抑制Tenascin-c/TLR4信号通路活化保护类风湿关节炎大鼠的炎症损伤。
Abstract:
Objective: To explore protective effects of resveratrol on inflammatory injury of the rats with rheumatoid arthritis (RA) and its influence on Tenascin-c/TLR4 signaling pathway. Methods: All 48 male SD rats were allocated to the control group, the model group, tripterygium glycoside (20 mg/kg) group, the groups of low, moderate and high doses of resveratrol (15, 30 and 60 mg/kg), eight rats each groups and they were drenched consecutively for 42 days. Except the control group, the rats in the other groups accepted multiple intradermal injection of bovine type Ⅱ collagen and complete Freund's adjuvant at the back and tail root of the rats at the first and 14th day of the experiment to prepare rat model with collagen induced arthritis (CIA). To record body weight, joint swelling and multiple arthritis indexes (AI) of the rats in different groups during therapeutic period, analyze the levels of IL-6, IL-1β and TNF-α, and measure the expressions of Tenascin-c and TLR4 protein in synovium tissue. Results: Compared with the control group, body mass of the rats grew slowly in the model group, hindlimb joints were swollen seriously, AI value increased signficantly, the levels of IL-6, IL-1β and TNF- α lifted notably, and the expressions of Tenascin-c and TLR4 protein increased remarkably (P<0.05); compared with the rats in the model groups, different intervention groups of medicine could improve body mass of RA rats, reduce joint swelling rate and AI values (P<0.05), they could inhibit the produce of the inflammatory factors including IL-6, IL-1β and TNF- α notably (P<0.05), and notably inhibit the expressions of Tenascin-c and TLR4 protein (P<0.05). Conclusion: Resveratrol could protect inflammatory injury of RA rats through inhibiting the activation of Tenascin-c/TLR4 signaling pathway.
