摘要:
目的研究仙茅-淫羊藿药对治疗卵巢早衰(premature ovarian failure,POF)的主要活性成分及其靶点的作用机制。 方法检索人类基因组注释数据库GeneCards,Drugbank数据库获取POF靶点,借助中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)获得仙茅、淫羊藿的潜在活性成分及其对应靶点,使用Bioinformatics & Evolutionary Genomics平台筛选仙茅-淫羊藿药对的活性成分与POF的共有靶点,利用Cytoscape软件构建网络图;采用String数据库构建蛋白质相互作用网络;使用DAVID数据库进行基因本体论富集分析(gene ontology,GO)与KEGG(kyoto encyclopedia of genes and genomes,KEGG)通路富集分析。 结果通过筛选得到30个药物活性成分,共涉及靶点278个,与疾病靶点有关的活性成分26个,得到仙茅、淫羊藿药对-疾病共同靶点41个,主要富集于139个生物过程和60条信号通路,涉及PI3K-Akt、甲状腺激素、MAPK等信号通路。 结论仙茅-淫羊藿药对活性靶点主要通过PI3K-Akt、甲状腺激素、MAPK等信号通路调控卵巢细胞过程及代谢发挥治疗作用。
Abstract:
ObjectiveTo study the principal active components and the targets of Xianmao (Curculiginis Rhizoma)-Yinyanghuo (Epimedii Folium) couplet medicines in the treatment of POF. MethodsPOF targets were obtained by searching GeneCards and Drugbank databases, the potential active ingredients and its corresponding targets of Xianmao and Yinyanghuo were gained via TCMSP, Bioinformatics&Evolutionary Genomics platforms were used to screen active ingredients of the couplet medicines and its common targets with POF, Cytoscape software was used to construct network planning; String database was adopted to build PPI; DAVID database was used to conduct GO enrichment and KEGG pathways enrichment analysis. ResultsThrough screening, there were 30 active ingredients, involving 278 targets, 26 active ingredients related to disease targets, as well as 41 common targets of Xianmao-Yinyanghuo couplet medicines and disease, primarily enriched in 139 biological processes and 60 signaling pathways, referring to PI3K-Akt signaling pathway, thyroid hormone signaling pathway, MAPK signaling pathway and others. ConclusionActive targets of Xianmao-Yinyanghuo couplet medicines develop therapeutic effects chiefly through regulating ovary cells and metabolism via PI3K-Akt signaling pathway, thyroid hormone signaling pathway, MAPK signaling pathway and others.
