摘要:
目的基于色差原理分析关黄柏的有效成分、浸出物含量与其外观颜色的相关性。 方法测定关黄柏的色差值及有效成分盐酸小檗碱、盐酸巴马汀及其浸出物含量,将代表颜色的外观指标值与代表含量指标值相关联,利用色差仪测定关黄柏色度,按照2020版《中华人民共和国药典》中关黄柏含量测定项及其浸出物测定项下方法,采用高效液相色谱仪及热浸法测定黄柏有效成分、浸出物含量;使用SPSS软件和SIMCA软件进行统计分析。 结果关黄柏中小檗碱及其浸出物含量与颜色的明亮度(L*)、黄蓝色(b*)、总色差值(E*ab)呈正相关,巴马汀含量与L*、a*呈正相关。 结论关黄柏外观颜色与其有效成分、浸出物含量相关,颜色越偏亮黄色,其有效成分及浸出物含量越高,质量相对较好;通过色差仪可以初步快速预测出关黄柏药材的有效成分及浸出物含量,可为关黄柏药材的质量评价提供参考。
Abstract:
ObjectiveTo analyze the correlation of the contents of active ingredients and the extracts of Guanghuangbo (phellodendri amurensis cortex), with its appearance and color based on the principle of color difference. MethodsAfter detecting the color difference values, the contents of the active ingredients such as berberine hydrochloride and bamatin hydrochloride, as well as the extracts of Guanghuangbo, the chromaticity of Guanghuangbo was measured by chromatometer when connecting the appearance indexes representing the color with the content indicators, according to the methods under the determination of content and extract of Guanghuangbo in 2020 edition of Chinese Pharmacopoeia, the contents of active ingredients and the extracts were detected by HPLC and hot water maceration; statistical analysis was carried out using SPSS software and SIMCA software. ResultsThe contents of berberine of Guanghuangbo and the extracts were positively correlated with L*, b* and E*ab of the color, the contents of bamatin were positively related to L* and a*. ConclusionThe appearance and color of Guanghuangbo are related to the contents of active ingredients and extracts of the materia medica, the brighter the color is, the higher the contents of active ingredients and the extracts are, the better the herb is; the contents of active ingredients and the extracts of the herb could be rapidly and primarilly determined by chromatometer, and it could provide the reference for quality evaluation of the herb.
