摘要:
目的研究槲皮素（quercetin，QUE）对胃癌的抑制及免疫调节作用。 方法通过CCK-8法（cell counting Kit-8，CCK-8）检测不同浓度QUE处理后HGC-27细胞存活率，筛选有效作用浓度；苏木精-伊红染色观察25 μM组、50 μM组处理24 h后HGC-27细胞形态；流式细胞术检测25 μM组、50 μM组处理24 h后细胞周期及凋亡变化；流式细胞术检测25 μM组、50 μM组处理后小鼠外周血中免疫细胞变化。 结果与对照组比较，25 μM组、50 μM组能够有效抑制HGC-27细胞增殖活性（P＜0.01）；50 μM组HGC-27细胞质空泡化，G0/G1期缩短（P＜0.001），S期延长（P＜0.001），G2/M期消失（P＜0.001）；50 μM组中细胞凋亡率增加（P＜0.001）；50 μM组小鼠外周血中T淋巴细胞比例增加（P＜0.001）。 结论QUE可以在S期阻滞HGC-27细胞增殖，促进细胞凋亡；其中高浓度QUE对于胃癌瘤体的抑制作用显著，QUE能够通过增强机体免疫功能发挥抑制肿瘤作用。
Abstract:
ObjectiveTo study the inhibitory effects of quercetin (QUE) on gastric cancer and its immuno-modulation mechanism. MethodsThe survival rate of HGC-27 cells after treatment with different concentrations of QUE was detected by CCK-8 to screen the effective concentrations; the morphology of HGC-27 cells was observed by HE staining method after 24 h of treatment in 25 μM and 50 μM groups; flow cytometry was used to detect the cell cycles and apoptosis after 24 h of treatment in 25 μM and 50 μM groups; and the changes in immune cells in the peripheral blood of mice after 24 h treatment in the 25 μM and 50 μM groups. ResultsCompared with the control group, 25 μM and 50 μM groups could effectively inhibit the activity of HGC-27 cells (P<0.01); HGC-27 cytoplasm became vacuolated after 50 μM QUE treatment, G0/G1 phase shortened (P<0.001), the S phase prolonged (P<0.001), the G2/M phase disappeared (P<0.001); cellular apoptosis increased remarkably in the 50 μM group (P<0.001); the increase in the proportion of T cells in the peripheral blood of the mice after 50 μM QUE treatment could be found (P<0.001). ConclusionQUE could block the proliferation of HGC-27 cells in the S phase and promote cellular apoptosis; among them, the high concentration of QUE has the significant inhibitory effects on the tumor of gastric cancer, QUE could inhibit tumor by enhancing the immune function of the body.
