摘要:
目的:建立蒙古扁桃叶中槲皮素含量测定的方法,对不同产地蒙古扁桃叶中槲皮素的含量进行比较。方法:采用HPLC法,Shim-Pack VP-ODS C18色谱柱(4.6 mm×250 mm,5 μm);流动相甲醇-0.2%磷酸(50∶50);柱温30℃;流速1.0 mL/min;检测波长360 nm;进样量20 μL。结果:线性关系良好,r=0.999 9,回归方程为Y=86130X-6471.9,线性范围为2.5~50 μg/mL,平均加样回收率为100.5%,RSD为1.9%(n=6)。不同产地蒙古扁桃叶中槲皮素的含量存在差异,内蒙古固阳地区的含量最高,贺兰山哈拉乌沟地区的含量最低。结论:本实验首次测定了蒙古扁桃叶中槲皮素的含量并对不同出产地进行比较,方法准确、可靠,可作为蒙古扁桃药材质量评价的指标及资源开发的依据。
Abstract:
Objective: To compare the contents of quercetin contained in Mongolian almond leaf [Amygdalus mongolica(Maxim.) Ricker] form different producing areas by establishing content determination method of quercetin. Methods: HPLC method was adopted, Shim-Pack VP-ODS C18 chromatographic column(4.6 mm×250 mm, 5 μm); mobile phase: methanol-0.2% phosphoric acid(50:50); column temperature 30℃; flow rate was 1.0mL/min; detection wavelength was 360 nm; the sample size was 20 μL. Results: Linear relationship was better, r=0.999 9, regression equation was Y=86130X-6471.9, linear range was from 2.5 to 50 μg/mL, average recovery rate was 100.5%, RSD was 1.9% (n=6). The difference was existed in the comparison of the contents of quercetin of Mongolian almond leaf from different producing areas, the contents of quercetin contained in Mongolian almond from Guyang district were the highest, while these contained in the Mongolian almond from Halawugou district of Helan mountains were the lowest. Conclusion: The contents of quercetin in Mongolian almond leaf are initially detected, and different producing areas are compared in the experiment, the method, accurate and reliable, could be taken as the index of quality evaluation of Mongolian almond and the reference of resource development.
