摘要:
目的利用网络药理学方法探究参苓白术散治疗功能性腹泻(functional diarrhea,FD)的作用机制。 方法在中药系统网络药理学分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)筛选参苓白术散中各中药的有效成分及相关作用靶点;在人类基因数据库(the human gene database,GeneCards)、PharmGKB数据库、DrugBank数据库、在线人类孟德尔遗传数据库(online mendelian inheritance in man,OMIM)及药物靶标数据库(therapeutic targetdatabase,TTD)中检索FD疾病相关靶点;使用R Studio 4.1.2对FD靶点及参苓白术散中的中药有效成分作用靶点取交集,使用STRING数据库对交集靶点进行蛋白-蛋白互作(protein-protein interactions,PPI)网络分析,结果导入Cytoscape 3.9.0中筛选关键靶点,然后对交集靶点进行基因本体论(gene ontology,GO)功能和京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路富集分析,并对关键靶点进行分子对接验证。 结果共筛选出160种中药有效成分,1936个靶点;疾病靶点7329个;参苓白术散中主要有效成分为刺槐素、山柰酚、木犀草素和槲皮素;关键靶点为TNF、CDK4、RB1、IL-6、CDK1、CDK2等;涉及脂质和动脉粥样硬化、PI3K/ATK等信号通路。 结论参苓白术散通过多成分、多靶点、多途径发挥治疗FD的作用。
Abstract:
ObjectiveTo explore the mechanism of Shenling Baizhu powder in the treatment of functional diarrhea (FD) using network pharmacology. MethodsThe active ingredients and the pertinent targets of herbs in Shenling Baizhu powder were screened from TCMSP, FD-related targets were searched from GeneCards, PharmGKB database, DrugBank database, OMIM and TTD; R Studio 4.1.2 was used to gain the intersection of FD targets and the targets of active ingredients of Shenling Baizhu powder, STRING database was applied to analyze PPI network of the intersected targets, the results were imported into Cytoscape 3.9.0 to screen for the key targets, and to perform GO and KEGG pathway enrichment analysis of the intersected targets, and to conduct molecular docking validation of the key targets. ResultsThe study has identified 160 active ingredients of the herbs, 1936 targets and 7329 disease targets; the main active ingredients of Shenling Baizhu powder contained acacetin, kaempferol, luteolin, and quercetin; the key targets were TNF, CDK4, RB1, IL-6, CDK1 and CDK2; and the study involved lipid and atherosclerosis, PI3K/AKT signaling pathway. Conclusion Shenling Baizhu powder could treat FD by multi-ingredient, multi-target and multi-pathway.
