摘要:
目的：观察养阴和胃方对糖尿病胃轻瘫大鼠的疗效及其对SCF/c-kit系统的影响。方法：将40只SD大鼠随机均分为正常对照组，模型对照组，养阴和胃方低、高剂量组，西药组，每组8只。采用腹腔注射注射用链脲佐菌素(STZ)建立糖尿病胃轻瘫大鼠模型，养阴和胃方低、高剂量组和西药组分别用低、高剂量养阴和胃方和吗丁啉混悬液灌胃。酚红灌胃实验测定胃排空率，Western Blot检测胃窦c-kit蛋白和清干细胞因子(stem cell factor，SCF)蛋白的表达，ELISA测定血清SCF含量。结果：与模型对照组相比，各给药组大鼠的胃排空率、c-kit蛋白、SCF蛋白和血清SCF含量均明显增高，差异有统计学意义(P＜0.05)，且随着中药浓度的增高，胃排空率、c-kit蛋白、SCF蛋白和血清SCF含量增高更加明显，差异有统计学意义(P＜0.05)。结论：中药养阴和胃方可上调SCF/c-kit系统的表达，从而逆转DGP胃排空障碍。
Abstract:
Objective: To observe the clinical effects of Yin-nourishing harmonizing-stomach prescription in treating the rats with diabetic gastroparesis (DGP) and investigate its influence on SCF/c-kit system. Methods: Forty SD rats were allocated to normal control group, model control group, western medicine group, low and high doses groups of Yin-nourishing harmonizing-stomach prescription randomly, eight rats each group. DGP rat model was established by intraperitoneal injection of STZ, low, high doses groups of Yin-nourishing harmonizing-stomach prescription and western medicine group were drenched with low, high doses of Yin-nourishing harmonizing-stomach prescription and motilium suspension. Gastric emptying rate was determined by phenol red gavage experiment, the expressions of c-kit and SCF proteins in gastric tissues were analyzed by Western blot, and the levels of SCF in the blood were detected by ELISA. Results: Compared with model control group, gastric emptying rate, the contents of c-kit protein, SCF protein and SCF in the serum raised obviously in the rats of different medication groups (P＜0.05), as the concentrations of herbs increased, gastric emptying rate, the contents of c-kit protein, SCF protein and SCF in the serum raised obviously, and the difference had statistical meaning(P＜0.05). Conclusion: Yin-nourishing harmonizing-stomach prescription could up regulate the expressions of SCF/c-kit system, therefore to reverse gastric emptying disorder of DGP.
