摘要:
目的:通过建立慢性萎缩性胃炎(CAG)大鼠模型,观察调胃饮对CAG大鼠血清胃泌素(GAS)和前列腺素E2(PGE2)的调控作用。方法:1)造模:选取体质量(200±20)g健康雄性SD大鼠35只,随机抽取6只作为空白对照组,剩余29只按综合法(饥饱失常结合氨水自由饮用,水杨酸钠及乙醇灌胃)建立CAG模型。2)分组:选取造模成功的大鼠24只,随机分为4组,模型组、维酶素组及调胃饮高、低剂量组。3)指标检测:给药8周后,观察各组大鼠体质量变化情况,检测血清GAS和PGE2水平,光镜下观察各组大鼠胃黏膜组织病理学改变情况。结果:1)干预前,空白对照组体质量各组大鼠大于其他各组(P<0.05),其他组之间比较差异无统计学意义(P>0.05);干预后,各组大鼠体质量均高于干预前(P<0.01),且空白对照组>调胃饮高剂量组>维酶素组>调胃饮低剂量组>模型组(P<0.05)。2)空白对照组血清GAS、PGE2均高于其他各组(P<0.05),且调胃饮高剂量组>维酶素组>调胃饮低剂量组>模型组(P<0.01)。3)空白对照组大鼠胃黏膜正常,模型组大鼠胃黏膜符合CAG的病理改变。各给药组与模型组相比,病理结果均有不同程度的改善,以调胃饮高剂量组为最佳。结论:调胃饮治疗CAG的机制可能与上调血清GAS和PGE2水平,提高胃黏膜保护能力,有效抵制有害物质对黏膜的损伤,增强胃黏膜血流量,促进胃黏膜的快速修复有关。
Abstract:
Objective: To observe the regulatory effects of TiaoWei drink on gastrin (GAS) and prostaglandin E2 (PGE2) of rat model by establishing the models with chronic atrophic gastritis (CAG). Methods: 1) modeling: 35 healthy SD rats, weighing(200±20)g, were chosen, six rats were randomly selected as blank control group, the remaining 29 rats were prepared into CAG models using synthetic methods (irregular diet combined with ammonia water, drink freely, intragastric administration of natrium salicylicum and ethanol). 2)grouping: 24 rat models were selected and divided into the model group, vatacoenayme group, high and low dose groups of TiaoWei drink. 3)detecting the indexes: after treating for eight weeks, to observe the changes of body weight of the rats in different groups, to detect the levels of GAS and PGE2, and to observe pathological changes of gastric mucosa under the light microscope. Results:1) Before the intervention, blank control group was larger than other groups in body mass significantly (P<0.05), the difference had no statistical meaning in the comparisons of other groups(P>0.05); after the intervention, body mass of other groups were higher than before the intervention(P<0.01), and blank control group>high dose group of Tiaowei drink>vatacoenayme group>low dose group of Tiaowei drink>the model group(P<0.05). 2) the levels of GAS and PGE2 of blank control group were higher than these of other groups(P<0.05), and high dose group of Tiaowei drink>vatacoenayme group>low dose group of Tiaowei drink>the model group(P<0.01). 3) gastric mucosa of the rats in blank control group was normal, while gastric mucosa of the rats in the model group was up to pathological changes of CAG. When different intervention groups were compared with the model group, pathological results improved to different degrees, high dose group of TiaoWei drink was the best. Conclusion: The mechanism of TiaoWei drink in the treatment for CAG might be related to up regulating the levels of GAS and PGE2 in the serum, raising protective capability to gastric mucosa, effectively resisting the damage of harmful substance to mucosa, enhancing blood flow of gastric mucosa and promoting rapid rehabilitation of gastric mucosa.
