目的:观察白杨素(Chrysin,ChR)对结肠癌SW480细胞体内外的抑制作用。方法:采用CCK-8法检测白杨素对SW480细胞增殖活力的影响,采用Hochest33258染色检测SW480细胞形态变化及凋亡,观察白杨素对体外结肠癌细胞的生长抑制作用;建立结肠癌SW480细胞株裸鼠异种移植瘤模型,共24只,分为4组,每组6只,即对照组、白杨素高、中、低剂量组。观察不同剂量白杨素对体内肿瘤生长的抑制作用。结果:白杨素对SW480细胞具有较强的增殖抑制作用,20、40、80 μM抑制率分别为10.10%、45.63%、61.96%,且呈剂量依赖关系;白杨素可诱导结肠癌SW480细胞凋亡,凋亡率高低呈剂量依赖关系;与对照组相比,白杨素各治疗组体内异种移植肿瘤的重量均减少(P<0.01),且白杨素低、中、高剂量组的抑瘤率分别为25.91%,39.98%和56.77%。结论:白杨素对体内外结肠癌SW480细胞生长具有较强的抑制作用,这与白杨素能够诱导其凋亡相关。
Abstract:
Objective: To observe the inhibitory effects of chrysin (CHR) on colon cancer SW480 cells in vitro and vivo. Methods: The effects of CHR on the proliferation of SW480 cells were detected by CCK-8 method, morphological changes and the apoptosis of SW480 cells by Hochest 33258 staining method, the inhibitory effects of CHR on colon cancer cells in vitro were observed; nude mice with xenograft transplated tumor of human colon cancer SW480 cell line were established, altogether 24 mice, they were randomized into four groups, six mice each group, that is, the control group, high, moderate and low dosages groups of CHR. The inhibitory effects of different dosages of CHR on the growth of the tumor in vitro were observed. Results: CHR showed stronger inhibitory effects to SW480 cells, the inhibitory rates at 20, 40 and 80 μM were 10.10%, 45.63% and 61.96% respectively, in a dose-dependent manner; CRH could induce the apoptosis of colon cancer SW480 cells, the apoptosis rate showed the dose-dependent relationship; compared with the control group, the weight of xenograft tumor in different treatment groups of CHR reduced (P<0.01), the tumor inhibition rates in low, moderate and high dosages groups of CHR were 25.91%, 39.98% and 56.77% respectively. Conclusion: CHR has stronger inhibitory effects on the growth of colon cancer SW480 cells in vitro and in vivo, and this might be related to inducing the apoptosis.
