摘要:
目的:探讨醒脑方对血管性痴呆(vascular dementia,VD)大鼠学习记忆能力及额叶、海马、胼胝体、视束组织学的影响。方法:将大鼠按随机数字表法随机分为假手术组、模型组、脑复康组及醒脑方小、中、大剂量组。采用双侧颈总动脉闭塞法建立大鼠慢性脑灌注不足诱发的VD模型。给药14天后,Morris水迷宫法检测大鼠学习记忆能力;HE染色观察大鼠额叶、海马CA1区组织病理学改变;LFB染色观察大鼠胼胝体、视束组织病理学改变。结果:与模型组比较,醒脑方0.84 g组第3、4天逃避潜伏期缩短(P<0.05),醒脑方0.42 g组第4天逃避潜伏期缩短(P<0.05),醒脑方高、中剂量组第4天逃避潜伏期缩短程度与脑复康组比较无明显差异(P>0.05)。此外,与模型组相比,醒脑方大、中、小剂量组穿台次数均增加(P<0.05),与脑复康组比较无明显差异(P>0.05)。醒脑方能改善大鼠海马CA1区神经元及胼胝体、视束组织病理学形态。结论:醒脑方对慢性脑灌注不足大鼠学习、记忆能力的改善作用与脑复康相当,其作用靶点可能与额叶、海马、胼胝体及视束有关。
Abstract:
Objective: To explore the effects of Xingnao prescription on learning and memory ability of brain tissue, the histology of frontal lobe, hippocampus, callosum and optic tract of the rats with vascular dementia (VD). Methods: All the rats were allocated to sham operation group, the model group, Naofukang group, low, moderate and high dose groups of Xingnao prescription. Bilateral common carotid artery occlusion was used to establish VD model induced by chronic cerebral hypoperfusion. After medicating for 14 days, Morris water maze was used to detect learning and memory ability of the rats; HE staining to observe histopathological changes of frontal lobe, hippocampal CA1 region; LFB staining to explore histopathological changes of callosum and optic tract of the rats.Results: Compared with the model group, the escape latency of the rats in Xingnao decoction group of 0.84g/kg shortened apparently at the 3rd and 4th day(P<0.05), the escape latency of the rats in Xingnao decoction group of 0.42g/kg shortened notably at the 4th day(P<0.05), there was no significant difference when high and moderate doses groups of Xingnao prescription were compared with Naofukang group in the shortening degree of the escape latency (P>0.05). Besides, compared with the model group, the times of rat crossing the platform increased notably in low, moderate and high dose groups of Xingnao prescription(P<0.05), it showed no obvious difference when it was compared with Naofukang group(P>0.05). Xingnao prescription could improve histopathological morphology of callosum, optic tract and neurons of hippocampal CA1 region.Conclusion: Xingnao prescription could improve learning and memory ability of brain tissue of VD rats, and its target might be related to frontal lobe, hippocampus, callosum and optic tract of the rats.
