摘要:
目的探讨加减薯蓣丸对APP/PS1小鼠海马区腺苷酸活化蛋白激酶(AMP-activated protein kinase,AMPK)、真核细胞延伸因子2激酶(Eukaryotic elongation factor 2 kinase,eEF2K)、真核细胞延伸因子2(Eukaryotic elongation factor 2,eEF2)蛋白水平的影响,探讨其参与改善AD病理的机制。 方法将5月龄雄性APP/PS1小鼠20只分为模型组和中药组,同窝野生型小鼠10只作为空白组,中药组每天予加减薯蓣丸14 g/kg,模型组及空白组予等体积生理盐水,灌胃28天。采用新物体识别实验测试小鼠学习记忆能力;蛋白免疫印迹法(Western Blot)检测小鼠海马区AMPKα1、eEF2K、eEF2对应磷酸化蛋白p-AMPKα1、p-eEF2K、p-eEF2、突触素(synapsin-1,SYN-1)以及突触后膜致密物95(postsynaptic density 95,PSD-95)的表达水平。 结果与空白组相比,模型组小鼠新物体识别能力下降;与模型组相比,中药组小鼠新物体识别能力上升。与空白组相比,模型组小鼠p-AMPKα1/AMPKα1、p-eEF2K/eEF2K、p-eEF2/eEF2不同程度上升,SYN-1、PSD-95表达下降,差异均具有统计学意义(P<0.05);与模型组相比,中药组小鼠p-AMPKα1/AMPKα1、p-eEF2K/eEF2K、p-eEF2/eEF2均下降,SYN-1、PSD-95表达上升,差异均具有统计学意义(P<0.05)。 结论加减薯蓣丸通过抑制APP/PS1小鼠海马区AMPKα1磷酸化水平,调节eEF2K/eEF2信号通路,影响AD的突触可塑性,以改善其病理状态,提高APP/PS1小鼠学习记忆能力。
Abstract:
ObjectiveTo explore the effects of modified yam pills on the levels of AMPK, eEF2K and eEF2 protein in the hippocampus of APP/PS1 mice, and discuss the mechanism of the medicine participating in the improvement of AD pathology. MethodsTwenty five-month male APP/PS1 mice were randomized into the model group and herbal group, ten wild mice in the same nest served as the blank group, herbal group took modified yam pills each day, 14 g/kg, the model group and the blank group were given the same volume of physiological saline by gavage for 28 days. New object recognition experiment was used to detect learning and memory ability of mice; Western blot method to detect the expressions of p-AMPKα1, p-eEF2K, p-eEF2, SYN-1 and PSD-95 in the hippocampus of APP/PS1 mice. ResultsCompared with the blank group, new object recognition ability of the mice in the model group lowered; compared with the model group, new object recognition ability of the mice of the herbal group lifted. Compared with the blank group, p-AMPKα1/AMPKα1, p-eEF2K/eEF2K and p-eEF2/eEF2 of the mice in the model group increased to different degrees, the expressions of SYN-1 and PSD-95 reduced, and the difference showed statistical meaning(P<0.05); compared with the model group, p-AMPKα1/AMPKα1, p-eEF2K/eEF2K and p-eEF2/eEF2 of the mice decreased in the herbal group, the expressions of SYN-1 and PSD-95 rose, and the difference was statistically significant(P<0.05). ConclusionModified yam pills could improve its pathological state and raise learning and memory ability of APP/PS1 mice through inhibiting phosphorylation level of AMPKα1 in the hippocampus of APP/PS1 mice, regulating eEF2K/eEF2 signaling pathway and affecting synaptic plasticity in AD.
